摘要
目的使用qRt-PCR技术,分析脱氢表雄酮对骨髓间充质干细胞成骨分化过程中相关成骨特征性基因表达的变化及其可能的机制。方法提取3~7天SD乳鼠干细胞,分为空白组和脱氢表雄酮组,分别处理4、7、14天后,采用茜素红验证干细胞成骨分化效果,提取各组总RNA行PCR检测相关成骨基因的表达。结果与空白组比较,脱氢表雄酮组在第14天表现出明显的矿化结节,其中成骨特征性基因Alp、Ocn和ColⅠ较对照组明显上调。结论脱氢表雄酮对干细胞成骨分化有明显促进作用,相关骨形成基因显著上调,并可能通过BMP/RUNX2构成的信号通路调节干细胞的成骨分化。
Objective The qRT-PCR technique was used to analyze the changes in the expression of genes related to osteogenic differentiation of BMSCs during osteogenic differentiation of bone marrow mesenchymal stem cells and possible mechanisms. Methods 3~7 days old SD rat neonatal rat stem cells were divided into blank group and dehydroepiandrosterone-treated group.After 4,7 and 14 days of treatment,alizarin red was used to verify the osteogenic differentiation of stem cells.Total RNA was extracted from each group to detection related osteogenic gene expression. Results Compared with the blank group,the DHEA induction group showed obvious mineralized nodules on the 14 th day,among which the osteogenic characteristic genes Alp,Ocn,and ColI were significantly up-regulated in the control group. Conclusion Dehydroepiandrosterone significantly promotes the osteogenic differentiation of stem cells,significantly up-regulates the related bone formation genes,and may regulate the osteogenic differentiation of stem cells through the signaling pathway composed of BMP/RUNX2.
