miRNA对C3H10T1/2细胞成软骨分化相关基因表达的影响

摘要 目的探讨miR-30c-5p介导的C3H10T1/2细胞成软骨分化相关基因表达的影响。方法取C3H10T1/2细胞系并转染miR-30c-5p后,RT-qPCR验证miR-30c-5p相关的成软骨基因。结果转染miR-30c-5p诱导第7、14、21天时,RT-qPCR检测软骨标志物acan、sox9、col2a1mRNA的基因表达均显著高于对照组和过表达组。结论抑制表达阳性组的miR-30c-5p促进了C3H10T1/2细胞的成软骨基因的表达。 Objective To investigate the effect of miR-30 c-5 p-mediated gene expression on chondrogenic differentiation of C3 H10 T1/2 cells. Methods The expression of acan,sox9,col2 a1 mRNA was detected by RT-qPCR on7,14,21 days after transfection with miR-30 c-5 p. Results The relative expression of acan,sox9,col2 a1 mRNA was significantly higher than that of the control and overexpression groups. Conclusion MiR-30 c-5 p inhibiting the expression of positive group promoted the expression of chondrogenic gene in C3 H10 T1/2 cells.
