同位素标记原儿茶酸-(^(13)COOH)的合成研究

摘要 设计了以3,4-二甲氧基溴苯为原料,经格利雅反应制备得到格式试剂,通入13 C标记的13 CO2气体,制备得中间体3,4-二甲氧基苯甲酸-(13 COOH)。将中间体用BBr3脱除甲基后得到粗产品,粗产品再经过活性炭脱色,冷却结晶得到原儿茶酸-(13 COOH)。设计的合成路线操作简单,工艺流程短,副产物少,收率可达45%,13 C同位素丰度稀释低。产物经HPLC、MS、1 H NMR和13 C NMR表征,结果表明:制备的原儿茶酸-(13 COOH)化学纯度>99%,13 C同位素丰度>98%,作为重要的示踪剂,可为医药和化工领域研究提供基础。 A novel synthesis method of protocatechuic acid-(13 COOH)was designed.3,4-dimethoxy bromobenzene as raw material,Grignard reagent was prepared by Grignard reaction,then passed through the 13 C labeled 13 CO2 gas,the intermediate 3,4-dimethoxy benzoic acid-(13 COOH)was prepared.The crude product was obtained viademethylation with BBr3,which was decolorizing by activated carbon.Protocatechuic acid-(13 COOH)was obtained by cooling crystallization.Synthesis route had the advantages of simple operation,short process flow,less side product,the yield was more than45%,13 C labeled isotope abundance was not diluted.The product was characterized by HPLC,MS,1 H NMR and 13 C NMR,the chemical purity was more than 99%,13 C enrichment was more than 98%.As an important tracer,it can provide a basis for medical and chemical research.
