乙酰氨基阿维菌素原位凝胶注射剂对小鼠的急性毒性研究

摘要 为了对乙酰氨基阿维菌素原位凝胶注射剂的安全性进行评价,采用简化寇氏法进行了小鼠的急性毒性研究。首先通过预试验的最小致死剂量(LD_(100))和最大耐受剂量(LD_0)确定给药剂量及试验分组。皮下注射给药,观察7 d,记录死亡数,计算LD_(50)及95%的置信区间。结果表明,乙酰氨基阿维菌素原位凝胶注射剂对小鼠的急性毒性LD_(50)为5.903 g/kg,95%置信区间为5.105 3~6.415 0 g/kg。按照化学药物剂量分级,乙酰氨基阿维菌素原位凝胶注射剂可认定为实际无毒物质,说明该药临床皮下注射使用安全。 An acute toxicity test of Eprinomectin in situ gel injection on mice was taken to evaluate its safety by simplified Karber's method. The dosage and grouping were determined based on the minimum lethal dose(LD(100)) and the maximum tolerated dose(LD0) in preliminary experiment. The mice were administered with Eprinomectin in situ gel injection and observed seven days. The number of deaths was recorded for calculating the LD(50)(lethal dose 50%) and the 95% confidence limit. The results showed that the acute toxicity LD(50) of Eprinomectin in situ gel injection on mice was 5.903 g/kg,95% confidence limit ranges was from 5.105 3 ~6.415 0 g/kg. It indicated that the Eprinomectin in situ gel injection was non-toxic substances according to chemically drug classification.
