补肺消积饮含药血清对人肺癌细胞A549的增殖抑制及凋亡诱导作用研究

Study of Medicated Serum of Bufei Xiaoji Decoction on the Role of Proliferation Inhibition and Apoptosis Induction in Human Lung Cancer A549 Cell Line

[目的]观察补肺消积饮含药血清对人肺癌细胞A549凋亡与增殖的影响。[方法]采用体外中药血清药理学方法,实验组分为8%补肺消积饮含药血清组与16%补肺消积饮含药血清组,设10%非含药血清为对照组。观察各组对人肺癌细胞A549的增殖抑制及凋亡诱导作用。[结果]8%和16%的补肺消积饮含药血清分别作用人肺癌A549细胞24 h后,细胞存活率分别为(83.5±0.66)%和(40.7±0.41)%,抑制率分别达到(16.5±0.34)%和(59.3±0.59)%;流式细胞仪检测8%与16%补肺消积饮含药血清作用24 h后A549细胞发生凋亡,凋亡率分别为(24.10±2.51)%、(35.20±4.42)%。其中16%补肺消积饮含药血清对A549细胞的增殖抑制及凋亡诱导作用优于8%补肺消积饮含药血清。[结论]在体外细胞实验中,补肺消积饮含药血清对人肺癌A549细胞的增殖有明显抑制作用,并可诱导人肺癌A549细胞凋亡。

[Objective] To observe the effect of medicated serum of Bufei Xiaoji decoction on proliferation inhibition and apoptosis induction in human lung cancer A549 cell line. [Methods] The experimental groups were divided into 8% medicated serum group and 16% medicated serum group according to the approach of serum pharmacology of Chinese medicinal in vitro. And 10% non-drug-containing serum was set as control group. The proliferation inhibition and apoptosis induction of human lung cancer A549 cell line was observed in each group. [Results] After treatment of medicated serum of Bufei Xiaoji decoction of 8% and 16% in human lung cancer A549 cell line for 24 hours, the cell survival rates were （83.5±0.66）% and （40.7±0.41）%, respectively,and the inhibition rates were （16.5±0.34）% and （59.3±0.59）%,respectively. The apoptosis of 549 cells was detected by flow cytometry after treatment of medicated serum of Bufei Xiooji decoction of 8% and 16% for 24 hours, and the apoptosis rates were （24.10±2.51） % and（35.20±4.42）%, respectively. The 16% medicated serum of Bufei Xiaoji decoction had better effect on proliferation inhibition and apoptosis induction in A549 cells than 8% medicated serum of Bufei Xiooji decoction. [Conclusion] In vitro cell experiments,medicated serum of Bufei Xiaoji decoction could significantly inhibit the proliferation and induce apoptosis in human lung cancer A549 cell line.
