二色补血草总黄酮对功能性子宫出血模型大鼠子宫内膜VEGF、MMP-9表达的影响 被引量:7

摘要 目的:观察二色补血草总黄酮对功能性子宫出血模型大鼠子宫内膜VEGF、MMP-9表达的影响。方法:大鼠去势并利用雌激素负荷法建立无排卵型功能失调性子宫出血(ADUB)子宫内膜增生症大鼠模型,给予二色补血草总黄酮后,观察大鼠子宫内膜血管内皮生长因子(VEGF)及基质金属蛋白酶-9(MMP-9)表达水平的变化。结果:造模后大鼠VEGF和MMP-9蛋白和m RNA表达水平均显著增加(P<0.01,P<0.05),导致子宫内膜异常增生。给药后,二色补血草总黄酮高剂量组可明显降低大鼠子宫内膜VEGF和MMP-9蛋白和m RNA表达水平(P<0.05);二色补血草总黄酮中剂量组能明显降低大鼠子宫内膜VEGF和MMP-9 m RNA表达水平(P<0.05)和VEGF蛋白水平(P<0.05)。结论:二色补血草总黄酮治疗ADUB的机制可能是通过下调VEGF的转录和翻译水平,引起MMP-9表达水平下降,减轻子宫内膜血管脆性从而达到止血的目的。 Objective: To observe the effect of total flavonoids from Limonium bicolor on the expressions of VEGF and MMP-9 in the endometrium of the rats with dysfunctional uterine bleeding. Methods: The rat model of ADUB hyperplasia endometrii was established by ovariectomy and estrogen load method. The expressions of VEGF and MMP-9 were observed in rats after the intervention of total flavonoids from Limonium bicolor. Results: The protein and m RNA expressions of VEGF and MMP-9 were significantly increased after modeling( P〈0. 01),which resluted in abnormal endometrial hyperplasia. The levels of VEGF and MMP-9 in the endometrium of the rats were significantly reduced after the intervention of medium-dose and high-dose of total flavonoids from Limonium bicolor( P〈0. 05). Conclusion: The mechansim of total flavonoids from Limonium bicolor treating ADUB may be down-regulating the transcription and translation of VEGF,resulting in the decrease of MMP-9 expression,thus to achieve the purpose of hemostasis by reducing the brittleness of endometrial vessels.
