摘要
目的建立同时测定银花感冒颗粒中绿原酸、甘草苷、5-O-甲基维斯阿米醇苷的含量的方法。方法色谱柱,Welch-C18色谱柱(4.6 mm×250 mm,5μm),流动相:乙腈(A)-0.4%磷酸(B),梯度洗脱0~5 min(5%~13%A);5~18 min(13%~14%A);18~22 min(14%~20%A);22~38 min(20%~32%A);流速1.0 mL·min^(-1);柱温30℃,检测波长327 nm,230 nm。结果银花感冒颗粒中绿原酸在54.05~540.50 ng(r=0.9999),甘草苷在35.58~355.80 ng(r=0.9993),5-O-甲基维斯阿米醇苷在30.59~305.90 ng(r=0.9999)范围内线性良好,平均回收率分别为99.58%(RSD为0.92%),99.28%(RSD为2.31%),98.91%(RSD为2.16%)。结论高效液相色谱波长切换法同时测定银花感冒颗粒中绿原酸、甘草苷、5-O-甲基维斯阿米醇苷的含量具有操作简单,专属性强,准确度高的优点,可以更全面的控制银花感冒颗粒的质量。
Objective A method was developed for simultaneous determination the contents of chlorogenic acid,Lquoritin and 5-Omethylvisammiosode in Yinhua Ganmao Granules by HPLC with wavelength switching method. Methods The chromatography separation was performed on the column of Agilent Welch-C18( 4. 6 mm × 250 mm,5 μm),with acetonitrile( A)-0. 4% phosphoric acid( B) as the mobile phase in the gradient elution mode( 0-5 min( 5%-13% A); 5-18 min( 13%-14% A); 18-22 min( 14%-20% A);22-38 min( 20%-32% A); at flow rate of 1. 0 mL·min^-1. The detection wavelength was 327 nm and 230 nm respectively,temperature of column was set at 30 ℃. Results The sample concentration of chlorogenic acid,Lquoritin and 5-O-methylvisammiosode had a good linearity with the value of peak areas at 54. 05-540. 50 ng( r = 0. 9999),35. 58-355. 80 ng( r = 0. 9993),30. 59-305. 90 ng( r= 0. 9999),respectively. The average recoveries were 99. 58%( RSD = 0. 92%),99. 28%( RSD = 2. 31%) and 98. 91%( RSD =2. 16%),respectively. Conclusions The method of simultaneous determination the contents of chlorogenic acid,Lquoritin and 5-Omethylvisammiosode in Yinhua Ganmao Granules by HPLC with wavelength switching method which has a good advantage of simple operation,high accuracy stability and can be used for the quality control of Yinhua Ganmao Granules effectively.
