摘要 目的检测微小RNA-103a(miR-103a)在宫颈癌细胞系中的表达水平,探讨其对宫颈癌细胞增殖、迁移和侵袭能力的影响。方法应用RT-qPCR法检测4种宫颈癌细胞株中miR-103a的表达水平;以宫颈癌细胞株HeLa为研究对象,分为miR-NC组(脂质体转染microRNA无关序列,即miR-NC)和miR-103a mimics组(脂质体转染miR-103a模拟物),用MTT法检测细胞增殖能力,划痕实验和Transwell实验评估miR-103a对宫颈癌细胞迁移和侵袭能力的影响。结果 miR-103a在宫颈癌细胞SiHa、HeLa、C33A和CaSki中均呈低表达,与对照细胞HaCaT中miR-103a的表达水平差异具有统计学意义(P<0.05);采用miR-103a mimics和miR-NC分别转染宫颈癌HeLa细胞后,与转染miR-NC的细胞相比,转染miR-103a的细胞增殖减少,迁移及侵袭能力受到明显抑制,其差异均具有统计学意义(均P<0.05)。结论 miR-103a在宫颈癌细胞系中呈低表达,上调miR-103a表达可减弱宫颈癌细胞增殖能力,并抑制细胞迁移及侵袭能力。 Objective To investigate the expression level of miR-103a in cervical cancer cell lines and explore its effect on the proliferation, migration and invasion of cervical cancer cells.Methods RT-qPCR was performed to detect the expression levels of miR-103a in four different cervical cancer cell lines. The HeLa cell lines were divided into miR-NC group(transfected with miR-NC) and miR-103a group(transfected with miR-103a mimics). MTT assay was used to detect the cell proliferation ability. Would healing and Transwell invasion assays were used to investigate the effect of miR-103a on migration and invasion of HeLa cells.Results The miR-103a was significantly down-regulated in cervical cancer cell lines( P 〈0.05). Furthermore, up-regulation of miR-103a in HeLa cells significantly reduced cell growth, migration and invasion( P 〈0.05).Conclusion The miR-103a could inhibit the proliferation, migration and invasion of cervical cancer cells.
