脂肪基质干细胞促进脂肪移植后血管化的研究 被引量:3

摘要 目的研究脂肪基质干细胞对脂肪移植物存活率及血管化的影响。方法体外分离培养兔脂肪组织中ADSCs,与自体兔脂肪颗粒混合移植于裸鼠背部。实验分为对照组(脂肪颗粒+生理盐水)和实验组(脂肪颗粒+ADSCs)。分别于移植后1,2,4,6周,通过B超检测和CD31免疫组化方法评价移植物体积和血管密度变化情况。结果脂肪组织中分离培养的ADSCs呈梭形生长,能够分化为成脂细胞和成骨细胞分化。脂肪移植后第1,2,4,6周,实验组脂肪移植物存活率为83.48%±4.91%,67.38%±8.04%,38.23%±5.99%,29.23%±3.61%;对照组存活率为82.98%±8.11%,62.86%±9.60%,21.39%±8.96%,14.29%±5.71%;实验组和对照组移植物存活率随时间逐渐降低,实验组脂肪移植物存活率高于对照组(P<0.05)。在第4周时,实验组与对照组脂肪移植物血管密度差异有统计学意义(P<0.05)。结论体外培养的脂肪基质干细胞移植物可促进脂肪移植物早期血管化,提高存活率。 Objective To investigate the effect of transplanted adipose derived mesenchymal stem cells( ADSCs) on the survival rate and vascularization of transplanted fat tissue. Methods ADSCs were isolated from fat tissue of rabbits,cultured in vitro and identified by differentiation of adipogenesis and osteogenesis to prove its multipotential capability. Then ADSCs were incorporated into transplanted fat tissue,and injected into the back of nude mice. The mice were divided into two groups: control group( fat granule + saline) and experimental group( fat granule + ADSCs). At week 1,2,4,6 after transplantation,B ultrasound was used to observe the volume change of fat graft,and CD31 immunohistochemistry was used to analyze micro-vascularization density( MVD). Results ADSCs were spindle-shaped,and had the ability of adipogenesis and osteogenesis. At week 1,2,4,6 after transplantation,the survival rate was83. 48% ±4. 91%,67. 38% ±8. 04%,38. 23% ±5. 99%,and 29. 23% ± 3. 61% in experimental group,respectively,and 82. 98% ±8. 11%,62. 86% ±9. 60%,21. 39% ±8. 96%,14. 29% ±5. 71% in control group. The survival rate of transplanted fat graft was higher in experimental group than in control group( P〈0. 05). At week 4,MVD had a significant difference between experimental group and control group( P〈0. 05). Conclusion ADSCs can increase the survival rate and early MVD of fat graft.
