马齿苋提取物外用对大鼠口腔溃疡模型的影响

Study on Pharmacodynamics of Portulaca Oleracea Extract in Rats Model with Oral Ulcer

目的:研究马齿苋提取物对创伤性和复发性口腔溃疡模型大鼠的作用,为马齿苋应用于抗口腔溃疡提供实验依据。方法:复制创伤性和复发性口腔溃疡两种大鼠模型,随机将造模大鼠分为模型对照组、阳性对照组以及马齿苋提取物高、中、低剂量组,另设健康大鼠为正常对照组,马齿苋提取物高、中、低剂量组分别给予马齿苋提取物(12%,6%,3%),阳性对照组给予口腔溃疡散(6%),模型对照组和正常对照组给予生理盐水,连续涂抹给药15 d,1次/天,测定并记录各组溃疡数量、直径、愈合时间和复发间隔时间,造模组织黏膜经HE染色后观察组织形态学变化,采用ELISA法检测血清中TNF-α、IL-8水平。结果:外用涂抹马齿苋提取物可显著减少创伤性和复发性口腔溃疡模型大鼠的溃疡形成数量,显著加速溃疡面愈合,并可显著延长复发性口腔溃疡的复发时间(P<0.05或P<0.01);其中马齿苋提取物高、中剂量组均能显著降低两种口腔溃疡模型大鼠血清中TNF-α、IL-8水平(P<0.05或P<0.01)。结论:马齿苋提取物对大鼠创伤性和复发性口腔溃疡有显著治疗作用,其机制可能与降低炎症反应中的TNF-α、IL-8水平有关。

To study the effect of Portulaca Oleracea extract on model rats with traumatic and recurrent oral ulcer,and to provide the experimental basis for the application of Portulaca Oleracea in treating oral ulcer. Methods：The model rats with traumatic and recurrent oral ulcer were randomly divided into the model control group, the positive control group, high, middle, and low dose groups of Portulaca Oleraeea Extract and another normal control group. The high, middle, and low dose groups of Portulaca Oleraeea Extract were treated with Portulaca Oleracea Extract （ 12%, 6%, 3% ）. Positive control group was administrated with oral ulcer powder. The control group and the normal control group were treated with normal saline. The administrations above were applied 15 days continuously, once a day. The number,diameter,healing time and interval time of ulcers in each group were measured and recorded. Morphological changes of the mucosa of the model tissue were observed by HE staining. The levels of TNF-α and IL-8 in serum were detected by ELISA. Results：Compared with those of the model group,the external application of Portulaca Oleraeea extract can significantly reduce the number of ulcers in model rats with traumatic and recurrent oral ulcer, accelerate the healing of ulcers, and prolong the recurrence time of recurrent oral ulcers （P〈0.05 or P〈0.01 ）. The levels of TNF-α and IL-8 in serum of both rats models of oral ulcer were significantly lowered （P〈0.05 or P〈0.01 ）.Conclusion：Portulaca Oleracea extract has significant therapeutic effect on traumatic and recurrent oral ulcer in rats The mechanism may be related to the reduction of TNF-c and IL-8 levels in inflammatory response.
