摘要
Yes相关蛋白(YAP)/TAZ同源蛋白作为Hippo信号通路中最主要的转录共激活因子,参与多种细胞的增殖、分化以及凋亡。近年来YAP/TAZ在肾脏疾病中的作用受到重视,其参与急性肾损伤的修复和慢性肾脏病的纤维化,在推动急性肾损伤向慢性肾脏病进展的过程中发挥重要的作用。YAP/TAZ激活状态下促增殖、抗凋亡的作用特点在多种肾脏疾病中成为研究的热点,并已逐步得到基础实验的支持。此外,YAP/TAZ抑制剂与激活剂的研发为肾脏疾病的发病机制研究以及治疗提供了新的方向。
Yes-associated protein(YAP) and its ortholog transcriptional co-activator with PDZ binding motif(TAZ),are critical transcriptional co-activators of the Hippo pathway,involved in the regulation of cell proliferation,differentiation and apoptosis.Recently,the study of YAP/TAZ in renal diseases has been received many attention.YAP/TAZ participates in the repair of acute renal injury and fibrosis of chronic renal disease,and plays an important role in the progression from acute kidney injury to chronic kidney disease.Its characteristics of promoting proliferation and anti-apoptosis under the activated state have become a research focus in many renal diseases,and been gradually supported by basic experiments.The development of inhibitors and its activators will provide a new research direction for the pathogenesis and treatment of renal diseases.
