换液频率对关节软骨体外保存效果的影响

摘要 探索组织培养法保存关节软骨方案中换液频率对于软骨体外保存效果的影响,为寻找最佳的骨软骨移植物保存方案提供实验依据。在猪膝关节股骨髁负重区取圆柱状骨软骨块60枚,随机分为3组:A组(不换液);B组(每7 d换液);C组(每2 d换液)。保存28 d后检测软骨细胞存活率、组织学(番红O染色)和生物力学性能(杨氏模量)。结果显示,第28 d时,各组软骨细胞存活率均出现降低,C组优于A组和B组(P<0.05);C组蛋白多糖含量高于A组和B组(P<0.05);C组杨氏模量高于A组和B组(P<0.05)。表明组织培养法保存关节软骨时,每2 d更换一次保存液的保存方案最优。提高换液频率可以维持保存液中的营养成分,为体外软骨细胞的代谢提供更充足的营养,提高关节软骨的体外保存效果。 To investigate the effect of the update frequency on the preservation of cartilage in the articular cartilage. Furthermore,to provide an effective experimental basis for the preservation of the osteochondral allograft. 60 pieces of cylindrical bone cartilage obtained from the lateral loading area of the femoral condyle from the pig knee joint in the aseptic conditions were randomly divided into 3 groups: group A( don't change liquid); Group B( exchange of liquid per 7 days); Group C( exchange of liquid per 2 days). The survival of chondrocytes,histological( Safranine O staining) and biomechanical properties were examined after 28 d preservation. In the 28 th d,the survival rate of chondrocytes in each group was decreased,the survival rate of chondrocytes in group C was better than group A and B( P 〈0. 05); About Proteoglycan content,group C was higher than group A and group B( P〈0. 05),group C was higher than group A and group B on Young's modulus( P 〈0. 05). It shows that it is better to replace the preservation solution every two days when use the tissue culture method to preserve articular cartilage. Increase the update frequency of the preservation solution,which can maintain the nutrient content in the preserved solution and provide more adequate nutrition for the osteochondral chondrocytes metabolism. Furthermore,it improves the preservation effect of the osteochondral allografts.
