黄花棘豆组培过程中愈伤组织诱导培养基的优化 被引量:1

摘要 为了诱导黄花棘豆组培过程中产生愈伤组织,本文以黄花棘豆种子发芽获得幼苗的叶片为外植体,采用正交试验,探讨在MS和1/2MS培养基上生长激素BAP、NAA和TDZ的不同浓度对黄花棘豆愈伤组织诱导的影响。结果表明:两种培养基上的种子发芽率差异不显著,均在70%以上。比较极差得到,在MS培养基中3种生长素对黄花棘豆组培过程中愈伤组织生长影响的主次顺序依次为:NAA>TDZ、BAP,在1/2MS培养基中的顺序为TDZ>NAA>BAP。由正交试验与数据分析可得在MS培养基中黄花棘豆出愈的最佳生长素配比为BAP浓度为1mg/L,NAA为1.6mg/L,TDZ为2mg/L,在1/2MS培养基中的最佳配比为BAP0.5mg/L,NAA 1.6mg/L,TDZ 2mg/L。 In order to induce the callus during the tissue culture of Oxytropisochrocephala, the effect of different concentrations of growth hormones BAP, NAA and TDZ on MS and 1/2MS medium on callus induction of Oxytropis ochrocephalaon was studied by orthogonal experiments, and the explants of Oxytropisochrocephala callus is obtained through the leaves of seedlings in our study. The results showed that there was no significant difference in seed germination rate between the MS and 1/2MS medium, both of which were above 70% . By comparison of the range differences, the effect of the three auxins on the callus growth in MS medium during the tissue culture of Oxytropisochrocephala was: NAA 〉TDZ, BAP, and the sequence in 1/2MS medium was TDZ 〉 NAA 〉 BAP. The optimum auxin ratio of BAP , NAA and TDZ was 1 mg/L ,1.6 mg/L, and 2 mg/L respectively in MS medium, and the optimal ratio was BAP 0. 5 mg/L, NAA 1.6 mg/L,and TDZ 2 mg/L in the 1/2MS medium according to orthogonal test and data analysis.
