高压氧预处理对大鼠脑出血后炎症反应的影响

摘要 目的观察高压氧预处理(hyperbaric oxygen preconditioning,HBOP)对大鼠脑出血(intracranial hemorrhage,ICH)后炎症反应的影响,探讨HBOP诱导大鼠ICH后神经保护的机制。方法选用72只雄性Spradgue-Dawley大鼠随机分为HBOP组和对照组各36只,HBOP组给予高压氧预处理,对照组给予假预处理。采用脑内自体动脉血注入法建立大鼠ICH模型,比较两组大鼠神经功能评分,血清IL-1β、IL-6的含量及炎症反应情况。结果 ICH建模48 h、72 h后,HBOP组大鼠神经功能评分均高于对照组,差异有统计学意义(P<0.05);ICH建模后72 h,HBOP组大鼠血清IL-1β和IL-6含量均低于对照组,差异有统计学意义(P<0.05);ICH建模后72 h,HBOP组大鼠血肿周围炎症细胞浸润程度轻于对照组。结论 HBOP可抑制大鼠ICH后IL-1β、IL-6的表达、抑制ICH后炎症反应,可能是HBOP减轻大鼠ICH后脑损伤的机制之一。 Objective To observe the effect of hyperbaric oxygen preconditioning( HBOP) on the inflammatory response after intracranial hemorrhage( ICH) in rats so as to study the mechanism of HBOP induced ICH neuroprotection in rats.Methods Seventy-two male Spradgue-Dawley rats were divided into the HBOP group( n = 36) and the control group( n =36). The HBOP group was given HBOP,the control group was given false preconditioning. ICH experimental model was established by intracerebral autologous arterial infusion. The neurological function score, serum IL-1β, IL-6 and inflammatory reaction were compared between the two groups. Results After modeling 48 h and 72 h,the neurological function score of HBOP group was higher than that of the control group( P〈0.05). The level of serum IL-1β and IL-6 in the HBOP group was lower than the control group( P〈0.05). After modeling 72 h,compared with the control group,the HBOP group had a significantly lower level of inflammatory cell infiltration. Conclusions HBOP can inhibit the expression of IL-1β and IL-6,the inflammatory reaction after ICH. It may be one of the mechanisms of HBOP alleviating brain damage after ICH in rats.
