摘要
目的探讨益生菌对人结肠癌SW-480细胞增殖的作用及其对Bcl-2和Bax表达的影响。方法采用四甲基偶氮唑盐(MTT)法观察嗜酸乳杆菌、乳双歧杆菌和长双歧杆菌三种益生菌成分(菌体、菌体壁和细胞质)对SW-480细胞增殖的作用,实时荧光定量聚合酶链反应和蛋白免疫印迹法分别检测三种益生菌细胞壁成分对SW-480细胞Bcl-2和Bax表达的影响。结果与对照组相比,三种益生菌成分(菌体、菌体壁和细胞质)处理48 h后,均可抑制SW-480细胞增殖;三种益生菌的细胞壁均抑制SW-480细胞Bcl-2表达;三种益生菌的细胞壁均促进SW-480细胞Bax表达,差异有统计学意义(P<0.05)。结论嗜酸乳杆菌、乳双歧杆菌和长双歧杆菌三种益生菌均可抑制SW-480细胞增殖,其机制可能与Bcl-2表达下调、Bax表达上调有关。
Objective To explore the effect of probitics on cell proliferation and the protein expression of Bcl-2 and Bax in human colon cancer SW-480 cells. Methods The growth inhibitory activities of Lactobacillus acidophilus,Bifidobacterium Lactis,and Bifidobacterium Longum( bacteria composition,cell wall composition and cytoplasmic composition) on SW-480 cells were evaluated by MTT assay. Western blot and qRT-PCR methods were used to measure the effect on the expression of Bcl-2 and Bax by cell wall composition of the three different probiotics respectively. Results Compared with the control,bacteria composition,cell wall composition and cytoplasmic composition of the three different probiotics inhibited the proliferation of SW-480 cells( P〈0.05),Bcl-2 expression was down-regulated by cell wall composition of the three probiotics( P〈0.05),Bax expression was up-regulated by cell wall composition of the three probiotics( P〈0.05).Conclusions The three probiotics of Lactobacillus acidophilus,Bifidobacterium Lactis,and Bifidobacterium Longum can inhibit the proliferation of SW-480 cells,and the mechanism may be related to the down-regulation of Bcl-2 expression andthe up-regulation of Bax expression.
