蛹虫草多糖的提取及抗氧化活性研究 被引量:19

摘要 [目的]研究蛹虫草多糖的提取及抗氧化活性。[方法]在比较几种常见的蛹虫草多糖提取方法的基础上,以蛹虫草多糖的得率为指标,采用正交试验法优化了蛹虫草多糖的提取工艺,并对蛹虫草多糖抗氧化活性进行了分析。[结果]热水浸提法的多糖提取率优于超声浸提法,蛹虫草多糖的最优提取工艺:料液比1∶20(g/m L),浸提温度80℃,浸提时间1.5 h,在此条件下蛹虫草多糖的提取率达到9.31%。在抗氧化活性方面,蛹虫草随着多糖浓度的增加,DPPH自由基清除活性和铁离子螯合能力均增加,在多糖浓度为4 mg/m L时,DPPH自由基清除率为38.69%,之后活性趋于平稳;在多糖浓度为3.5 mg/m L时,其铁离子螯合率为56.66%,此后铁离子螯合能力亦保持平稳。[结论]该研究可为蛹虫草多糖的进一步分离纯化、活性研究及开发利用奠定基础。 [Objective]To study the extraction and antioxidant activity of polysaccharide from Cordyceps militaris.[Method]Based on the comparison of several common extraction methods of Cordyceps militaris polysaccharides,and taking the yield of Cordyceps militaris polysaccharide as an index,the extraction technology of polysaccharide from Cordyceps militaris was optimized by orthogonal experiment,and the antioxidant activity of Cordyceps militaris polysaccharides was analyzed. [Result]The experimental results showed that the extraction rate of polysaccharides from hot water extraction was better than that of ultrasonic extraction. The optimum extraction technology of Cordyceps militaris polysaccharide was: the ratio of material to liquid was 1 ∶20( g/m L),the extraction temperature was 80 ℃,and the extraction time was 1. 5 h. Under these conditions,the extraction rate of polysaccharide from Cordyceps militaris reached 9. 31%. The antioxidant activity of Cordyceps polysaccharide,with the increase of concentration of the DPPH radical scavenging activity and iron ion chelating ability were increased,the concentration of polysaccharide was4 mg/m L,DPPH free radical scavenging rate was 38. 69%,then the activity tended to be stable; when the polysaccharide concentration was3. 5 mg/m L,the rate of iron ion was 56. 66%,then the iron ion chelating ability also remained stable.[Conclusion]This study can lay a foundation for further isolation,purification,activity research and development of Cordyceps militaris polysaccharides.
