摘要 目的观察不同时点血吸虫肝损伤模型小鼠,探讨IL-17与肉芽肿病变及血清学改变的相关性。方法将50只昆明小鼠随机分为正常对照组和感染组,各25只。感染组小鼠人工感染日本血吸虫尾蚴,分别于感染后第14、28、42、56、70 d剖杀,每次5只,正常对照组剖杀时间点与感染组一致,测定血清谷丙转氨酶(GPT)、谷草转氨酶(GOT);取肝组织,肉眼观察肝脏表面肉芽肿炎症动态变化;取肝脏约0.5 g,匀浆,收集匀浆液上清,ELISA检测肝脏匀浆上清IL-17动态变化。结果大体病理标本肉眼观显示小鼠肝脏肉芽肿炎症在42 d左右最明显;血清学结果显示GPT/GOT在感染后42 d达到最高值。感染后28 d,肝脏局部匀浆上清中IL-17开始明显增加,42~56 d达到高峰,随后下降。结论小鼠感染日本血吸虫后,肝脏肉芽肿的严重程度、肝脏的损伤与IL-17蛋白水平的变化可能有相关性。 Objective To observe the hepatic injury model mice induced by schistosomiasis japonicum at different time points in order to investigate the correlation among granulomatous lesions and serological indexes. Methods 50 Kunming mice were randomly divided to normal control group and infection group,with 25 in each group.The mice in infection group were artificially infected with the Schistosoma japonicum cercaria and were dissected at 14,28,42,56,70 d after infection,5 at a time. The time point of dissection in the normal control group was the same as that in the infection group.Then the serum glutamic-pyruvic transaminase( GPT) and the glutamic-oxaloacetic transaminase( GOT) were measured; the dynamic changes of granuloma inflammation on the surface of liver were observed by taking liver tissue; about 0.5 g of liver was taken and was in homogenate,which was collected in supernatant,and dynamic changes of il-17 in liver homogenate were detected by ELISA. Results Macroscopic observation of the gross pathological specimen showed that the inflammation of hepatic granuloma was the most obvious at around 42 days. Serological results showed that GPT/GOT peaked at 42 d after infection. Il-17 in liver local homogenate supernatant began to increase significantly at 28 d after infection,peaked at 42-56 d and then decreased. Conclusion The severity and injury of hepatic granuloma in mice infected with schistosomiasis japonicum may be related to changes in il-17 protein levels.
