摘要
目的观察姜黄素对胰腺癌细胞应用吉西他滨化疗后增殖及凋亡的影响。方法通过对不同恶性程度的胰腺癌细胞株BxPC3、PANC-1进行细胞培养,将两种细胞分为对照组(A组)、吉西他滨治疗组(B组)、姜黄素治疗组(C组)、姜黄素联合吉西他滨治疗组(D组),采用噻唑蓝(MTT)法检测细胞增殖,膜联蛋白V-异硫氰酸荧光素(Annexin V-FITC)/碘化丙锭(PI)双染法检测细胞凋亡,PI单染色法检测细胞周期,明确药物阻滞细胞周期。结果BxPC3细胞单独应用吉西他滨化疗与联合姜黄素治疗比较,细胞活力由(83.27±4.13)%下降至(8.12±0.37)%(P=0.007),细胞凋亡率由(6.73±2.19)%增加至(26.33±3.71)%(P=0.006);PANC-1细胞单独应用吉西他滨化疗与联合姜黄素治疗比较,细胞活力由(83.77±6.03)%下降至(7.58±1.13)%(P=0.008),细胞凋亡率由(6.21±1.23)%增加至(25.08±3.24)%(P=0.005)。结论姜黄素联合吉西他滨可明显抑制胰腺癌细胞增殖,促进其凋亡。
ObjectiveTo investigate the effect of curcumin on proliferation and apoptosis of pancreatic cancer cells treated with gemcitabine chemotherapy.MethodsCell culture of pancreatic cancer cell lines BxPC3 and PANC-1 with different degrees of malignancy were performed. The two cells were divided into control groups. (Group A), gemcitabine treatment group (group B), curcumin treatment group (group C), curcumin plus gemcitabine treatment group (group D), methyl thiazol tetrazolium (MTT) was used to detect cell proliferation, and Annexin V-FITC/propidium iodide (PI) double staining The method was used to detect the apoptosis of cells. The PI staining method was used to detect the cell cycle and the drug was used to block the cell cycle.ResultsCompared with curcumin alone, the viability of BxPC3 cells treated with gemcitabine alone was decreased from (83.27±4.13)% to (8.12±0.37)% (P=0.007), and the apoptosis rate was (6.73±2.19)%. Increased to (26.33±3.71)% (P=0.006); cell viability decreased from (83.77±6.03)% to (7.58±1.13)% (P=0.008) in PANC-1 cells treated with gemcitabine alone and curcumin alone. The apoptosis rate increased from (6.21±1.23)% to (25.08±3.24)% (P=0.005).ConclusionCurcumin combined with gemcitabine can significantly inhibit the proliferation of pancreatic cancer cells and promote their apoptosis, which provides a new idea for the treatment of pancreatic cancer.
