Expression and clinical significance of short-chain dehydrogenase/reductase 16C5 in human pancreatic cancer tissues and cell lines

摘要 目的探讨短链脱氢还原酶16C成员5(SDR16C5)在胰腺癌中的表达及其与胰十二指肠切除术预后的关系。方法免疫组织化学检测SDR16C5在100例人胰腺癌组织及30例癌旁组织中的表达;Western blot及实时荧光定量聚合酶链反应(FQ-PCR)检测SDR16C5在人胰腺癌细胞株及人永生化胰腺导管上皮细胞株中的表达。Kaplan-Meier及COX回归分析SDR16C5表达对胰十二指肠切除术预后的影响。结果SDR16C5的表达与胰腺癌患者的性别、年龄、肿瘤直径、肿瘤部位、有无门脉侵犯无关。SDR16C5在胰腺癌组织及胰腺癌细胞株中的表达显著高于癌旁组织及永生化胰腺导管上皮细胞(χ^2=78.87,P=0.001)。低分化胰腺癌患者组织中SDR16C5的表达显著高于中、高分化胰腺癌(χ^2=42.46,P=0.001),伴有局部淋巴结转移者SDR16C5表达显著高于无局部淋巴结转移患者(χ^2=8.94,P=0.003),TNM分期Ⅲ期患者SDR16C5表达显著高于TNMⅠ、Ⅱ期患者(χ^2=8.94,P=0.003),SDR16C5中、高表达患者术后早期复发转移率显著高于低表达者(χ^2=39.02,P=0.001)。Kaplan-Meier法分析显示SDR16C5中、高表达患者胰十二指肠切除术后累计无病生存率(χ^2=6.29,P=0.012)及总体生存率(χ^2=11.63,P=0.001)明显低于低表达者,多因素COX回归分析显示SDR16C5是影响胰十二指肠切除术预后的独立危险因素。结论SDR16C5在胰腺癌组织及胰腺癌细胞株中的表达上调,检测SDR16C5有助于判断预后。 ObjectiveBy studying the expression level of short-chain dehydrogenase/reductase 16C5 (SDR16C5) in human pancreatic cancer to determine whether SDR16C5 is associated with the prognosis of pancreatoduodenectomy.MethodsImmunohistochemistery staining was used to detect the expression level of SDR16C5 in 100 human pancreatic cancer tissues and 30 paracancer tissues. Western blotting and real-time fluorescent quantitative polymerase chain reaction (FQ-PCR) was used to detect the protein and mRNA expression level of SDR16C5 in pancreatic cancer cell lines and immortalized pancreatic ductal epithelial cell line. Kaplan-Meier method and multivariate COX regression analysis was used to analyze the effect of SDR16C5 expression on the prognosis of pancreatoduodenectomy.ResultsThe expression level of SDR16C5 was not associated with gender, age, tumor diameter, tumor location and portal vein invasion in pancreatic cancer patients. The expression level of SDR16C5 in pancreatic cancer tissues and pancreatic cancer cell lines was significantly higher than in adjacent tissues and immortalized pancreatic ductal epithelial cell lines (P=0.001). The expression level of SDR16C5 in patients with poorly differentiated pancreatic cancer was significantly higher than that in medium and well differentiated pancreatic cancer (χ^2=42.46, P=0.001). SDR16C5 expression was significantly higher in patients with local lymph node metastasis than in patients without local lymph node metastasis (χ^2=8.94, P=0.003). The expression of SDR16C5 in TNM stage Ⅲ patients was significantly higher than that in TNMⅠ and Ⅱ patients (χ^2=8.94, P=0.003). The recurrence rate of those medium and high expression of SDR16C5 was significantly higher than in low expression patients (χ^2=39.02, P=0.001). The cumulative disease-free (χ^2=6.29, P=0.012) and overall (χ^2=11.63, P=0.001) survival rate of these patients with high expression of SDR16C5 was significantly lower than that of low expression. Multivariate COX regression analysis showed that SDR16C5 was an independent risk factor for the prognosis of pancreaticoduodenectomy.ConclusionSDR16C5 is up-regulated in pancreatic cancer tissues and pancreatic cancer cell lines. Detection of SDR16C5 helps to determine the prognosis.
