摘要
目的观察人类快速延迟整流性钾通道基因(HERG1)表达的HERG1蛋白和基质金属蛋白酶(MMP)-9在胃癌组织中的表达和相关性,探讨两者与胃癌预后的关系。方法应用免疫组织化学法[链霉菌抗生物素蛋白-过氧化物酶(SP)法]、反转录-聚合酶链反应(RT-qPCR)技术及Western blot方法检测80例胃癌组织标本及对应正常胃黏膜组织中的HERG1蛋白和MMP-9的表达,并进行回顾性随访。结果免疫组织化学、RT-qPCR、Western blot分析HERG1、MMP-9在胃癌组织中的阳性表达率分别为55%,60%;HERG1蛋白表达与淋巴结转移,远处转移关系密切;MMP-9的表达与胃癌的浸润程度、TNM分期、分化程度、淋巴结转移明显相关(P=0.000),而与性别、肿瘤大小、部位无明显相关。结论HERG1蛋白和MMP-9的过度表达与胃癌的发生发展有关,在肿瘤侵袭、转移过程中起重要作用。
ObjectiveTo investigate the expression of human ether-a-go-go-related gene (HERG1) protein and matrix metalloproteinase-9 (MMP-9) in gastric carcinoma and their correlation with prognosis of gastric carcinoma.MethodsReverse transcription and polymerase chain reaction (PCR) assay and Western blotting were used to detect the expression of HERG1 and MMP-9 in 80 cases of gastric carcinomas and corresponding normal gastric tissues, and the patients were followed up.ResultsThe positive expression rate of HERG1 and MMP-9 was 60% and 55% respectively. There was significant correlation between the positive expression rate of MMP-9 with depth of invasion, TNM stage, differention, lymphatic and distant matastasis. The positive rate of HERG1 was correlated with lymphatic and distant metastasis.ConclusionHERG1 protein and MMP-9 expression in gastric carcinoma appears to be well-characterized indicators of prognosis and metastasis and may play an important role in the tumor invasion and metastasis.
