摘要
目的构建稳定的SD大鼠脊髓损伤(SCI)模型,为SCI新治疗措施提供标准的疾病模型。方法雄性SD大鼠60只,随机分为模型组(A组)和假手术组(B组),每组30只,模型组采用Hatteras Instruments PCI3000精密打击器建立T10节段的SD大鼠SCI模型。于术后1、3、7、14、28 d每组分别取6只大鼠,采用运动功能评分及脚印分析(BBB)评分标准评价双后肢运动功能后进行脊髓组织学检测,观察细胞凋亡率和NF-200的表达量。结果模型组BBB评分较术前明显下降,术后各时间点均明显低于假手术组(P=0.000);假手术组凋亡指数在术后各时间点均明显低于模型组(P=0.000),而NF-200的表达显著高于模型组(P=0.000)。结论Hatteras Instruments PCI3000精密打击器撞击力度客观准确,打击精准,重复性好,是较为理想的SCI模型制备仪器。
ObjectiveTo establish reliable spinal cord injury (SCI) models in rodents for pathological/physiological changes after spinal cord injury to formulate standard models and provide reasonable prospects for new treatment strategy.MethodsSixty female SD rodents were randomly divided into model group and fake-operation group. In model group, SCI models were established in SD rodents at T10 segment via Hatteras PCI3000 instruments. At 1st, 3rd, 7th, 14th and 28th days after operation, spinal tissue analysis for neural necrosis rate and NF-200 expression were performed and analyzed after evaluating movement of posterior limbs by basso beattie bresnahan (BBB)scale in 6 rodents from 2 groups.ResultsIn model group, BBB scale decreased greatly after operaton along with inferiority in comparison with fake-operation group (P=0.000). In fake-opeation group, necrosis index was inferior yet NF-200 expression was higher than model group (P=0.000).ConclusionHatteras PCI3000 instruments contributes to establishing reliable SCI models by accurate and metrizable, controllable and repeatable punching.
