摘要
目的通过双指标优化全蝎白术混合发酵品水提工艺并探究其体外抗肿瘤活性。方法通过超声提取法,以总多糖及总蛋白得率为指标,分别考察超声时间、p H及料液比对提取率的影响。试验在单因素试验基础上,综合考虑双指标,通过正交试验法优化混合发酵品水提工艺。在最佳提取工艺条件下,采用MTT法进行体外抗肿瘤试验。结果最优工艺条件为:超声时间60 min、p H=11及料液比1∶60。在此条件下,全蝎白术混合发酵品中总多糖得率为41.316%,总蛋白得率为16.618%,综合评分达到最高。体外抗肿瘤结果显示发酵后效果更显著。结论通过双指标优化全蝎白术混合发酵品水提工艺条件,可以同时兼顾蛋白及多糖两种抗肿瘤有效成分,获得具有优良抗肿瘤活性的水提液,可为后续抗肿瘤试验的深入研究提供基础。
Objective To optimize water extraction of mixed fermentation products of scorpion and atractylodes with double indexes and determine its anti-tumor activity in vitro. Methods With the extraction rate of total polysaccharide and total protein as indexes, the effects of ultrasonic time, pH, and the ratio of materials on ultrasonic extraction were investigated. On the basis of single factor test and the double indexes water extracting process was optimized. Under the optimum extraction conditions, MTT was used to test the anti-tumor activity in vitro. Results The optimized process was as follows: ultrasonic time 60 rain, pH at 11, and 1 : 60 for the ratio of materials. The total polysaccharide was 41.316%, and the total protein yield was 16.618%, with the highest overall score. The anti-tumor effect in vitro was obvious after fermentation. Conclusion The double index optimization of water extraction process for mixed fermentation products of scorpion and atractylodes can determine both two anti-cancer active ingredients of protein and polysaccharide. The water extraction has excellent anti-tumor activity. This experiment provides the base for further anti-tumor research.
