MUC1和MUC5AC表达水平与食管癌预后的相关性 被引量:4

Expressions and clinical significances of MUC1 and MUC5AC in esophageal cancer

摘要 目的探讨黏蛋白1(MUC1)和黏蛋白5AC(MUC5AC)在食管癌中的表达及临床意义。方法选择2014年11月至2017年2月在本院进行肿瘤切除手术的食管癌患者51例作为研究对象,其中38例患者癌旁存在正常食管组织,分别分离患者肿瘤组织和癌旁正常组织。采用蛋白质免疫印迹和组织化学染色法分别检测MUC1和MUC5AC在肿瘤组织和正常组织中的表达水平,分析肿瘤组织中MUC1和MUC5AC表达水平与食管癌患者临床病理特征及预后的关系。结果 MUC1主要在肿瘤组织细胞核表达,在正常组织细胞膜和少量细胞核表达,其在肿瘤组织中相对表达量和阳性表达率均显著高于正常组织(P<0.05);MUC5AC主要在正常组织细胞膜表达,在肿瘤组织细胞质微弱表达,其在肿瘤组织中相对表达量和阳性表达率均显著低于正常组织(P<0.05);患者MUC1表达在T分期、淋巴结是否转移、肿瘤分化程度方面差异有统计学意义(P<0.05),MUC5AC表达在淋巴结是否转移方面差异有统计学意义(P<0.05);MUC1阳性表达患者5年死亡率显著高于阴性表达患者(P<0.05),MUC5AC阳性表达患者5年死亡率显著低于阴性表达患者(P<0.05);淋巴结转移、肿瘤分化程度、MUC1阳性表达和MUC5AC阴性表达是影响患者预后的危险因素。结论食管癌患者肿瘤组织中MUC1和MUC5AC表达水平与患者预后有关,可作为评估患者预后的生物学指标。 Objective To investigate the expressions and clinical significances of MUC1 and MUC5 AC in esophagealcancer.Methods Fifty-one cases of esophageal cancer patients who underwent tumor resection in our hospital fromNovember 2014 to February 2017 were selected as the subjects;among them,there were thirty-eight patients with normalesophageal tissues near the cancer,the tumor tissues and the precancerous normal tissues were isolated.Western blot andhistochemical staining were used to detect the expression levels of MUC1 and MUC5 AC in tumor tissues and normal tissues.The relationships between the expression levels of MUC1 and MUC5 AC in tumor tissues with the clinicopathological featuresand prognosis of the patients with esophageal cancer was analyzed.Results The MUC1 was mainly expressed in the cellnucleus of the tumor tissues,and was a little expressed in the cell membrane and nucleus of the normal tissues;the relativeexpression and the positive expression of MUC1 rate in tumor tissues was significantly higher than that in normal tissues(P〈0.05).MUC5 AC was mainly expressed in the cell membrane of normal tissues,weakly expressed in the cytoplasm of thetumor tissues;the relative expression and the positive expression rate of MUC5 AC in tumor tissues was significantly lowerthan that in normal tissues(P〈0.05).There was significant difference in the MUC1 expressions between the T staging,lymph node metastasis and tumor differentiation degree(P〈0.05);the differences of MUC5 AC expression in lymph nodemetastasis was statistically significant(P〈0.05).The 5 years' mortality rate of MUC1 positive expression patients wassignificantly higher than that of negative expression patients(P〈0.05).The 5 years' mortality rate of MUC5 AC positiveexpression patients was significantly lower than that of negative expression patients(P〈0.05);lymph node metastasis,tumor differentiation,MUC1 positive expression and MUC5 AC negative expression were the risk factors that affected theprognosis of the patients.Conclusion The expression levels of MUC1 and MUC5 AC in the tumor tissues of the patientswith esophageal cancer was correlated with the prognosis of the patients,which could be used as a biological indicator toevaluate the prognosis of the patients.
