耐力运动对营养性肥胖大鼠Galectin-3表达的影响 被引量:5

摘要 目的:探讨耐力运动能否通过影响半乳糖凝集素3(Galectin-3)的表达,进而改善营养性肥胖大鼠的胰岛素抵抗状况。方法:6周龄雄性SD大鼠30只,随机分成普通饲料喂养组(N组,n=8)和高脂饲料喂养组(HFD组,n=22),HFD组进行为期8周的肥胖大鼠造模,造模成功后又随机分成安静对照组(C组,n=8)和耐力运动组(E组,n=8),E组进行8周的中等强度耐力训练,末次训练后,N、C和E组大鼠采集内脏脂肪、腹主动脉取血并进行相关指标测定。结果:8周高脂饲料喂养后,HFD组大鼠体重和Lee’s指数显著高于N组(P<0.05),HFD组大鼠血清甘油三酯、总胆固醇、低密度脂蛋白胆固醇以及血糖水平显著高于N组,而高密度脂蛋白胆固醇则显著降低(P<0.05)。8周运动干预后,C组大鼠体重、内脏脂肪百分比、Gal-3、瘦素(Leptin)水平显著高于N组(P<0.05);E组大鼠的内脏脂肪百分比、Gal-3、Leptin浓度和胰岛素抵抗指数(HOMA-IR)指数均显著低于C组(P<0.05),血脂四项指标也有显著性差异(P<0.05);另外,Gal-3的浓度与体重、Leptin浓度和HOMA-IR指数均存在显著性相关。结论:高脂喂养的肥胖大鼠血脂代谢紊乱,Gal-3浓度升高,8周的耐力运动可能通过降低血液中Gal-3的浓度改善肥胖大鼠的胰岛素抵抗。 Objective To explore if endurance exercise can improve the insulin resistance(IR) of di-et-induced obese rats by affecting the expression of galectin-3(Gal-3). Method Thirty 6-week-oldmale Sprague-Dawley rats were randomly divided into a normal diet feeding group(group N,n=8) anda high fat diet group(group HFD,n=22). The HFD rats became obese after 8 weeks' high-diet feed-ing,and were further divided into a sedentary control group(group C,n=8) and an endurance exercisegroup(group E,n=8) randomly. Group E was subjected to moderate-intensity endurance training for 8 weeks. After the training,the visceral fat and the blood of abdominal aorta were collected from the ratsof group N,C and E and evaluated. Results After 8 weeks of high-fat diet feeding,the body weight,Lee's index,level of triglyceride,total cholesterol,low-density lipoprotein cholesterol and blood glucoseof the rats in the HFD group were significantly higher than those in the N group(P〈0.05),while thelevel of high-density lipoprotein cholesterol of the former group was significantly lower than the lattergroup(P〈0.05). After the training of 8 weeks,the level of body weight,visceral fat percentage(VFR),Gal-3 and leptin of the rats in group C were significantly higher than those of group N(P〈0.05),while the level of VFR,Gal-3,leptin and the homeostasis model assessment of insulin resistance(HOMA-IR) index of rats in group E were significantly lower than those in group C(P〈0.05),andthere were significant differences in the four indexes of blood lipids between group C and E(P〈0.05).Moreover,the expression of Gal-3 was significantly correlated with the body weight,concentration ofleptin and HOMA-IR index. Conclusion The diet-induced obese rats display a disorder of blood lip-ids and increased expression of Gal-3,and 8-week endurance exercise may improve their insulin resis-tance by decreasing the expression of Gal-3 in their blood.
