酸解氧化魔芋葡甘露聚糖对齐口裂腹鱼肠道免疫功能的影响 被引量:1

Effects of Dietary Acidolysis Oxidized of Konjac Glucomannan on Intestinal Immune Function in Schizothorax prenanti

摘要 在日粮中添加酸解氧化魔芋葡甘露聚糖1.6%(acidolysis oxidized of konjacgluconmannan,A-OKGM),饲养齐口裂腹鱼(Schizothorax prenanti)60 d后,齐口裂腹鱼经腹腔注射9伊108cfu/m L的嗜水气单胞菌(Aeromonas hydrophila,Ah)0.2 mL,在注射嗜水气单胞菌1周后,分别取齐口裂腹鱼肠道分成前肠、中肠和后肠段。利用酶标仪测定溶菌酶(LZM)和酸性磷酸酶(ACP);并运用荧光定量PCR分析酸解氧化魔芋葡甘露聚糖对齐口裂腹鱼感染嗜水气单胞菌后不同肠段中白细胞介素-1β(il-1β)、白细胞介素-10(il-10)、白细胞介素-8(il-8)、肿瘤坏死因子(tnf-α)和转化生长因子β(tgf-β1)的表达影响。结果表明,与注射磷酸盐缓冲液对照组相比,感染嗜水气单胞菌A-OKGM组对齐口裂腹鱼的前肠、中肠、后肠中溶菌酶的活性有增加的趋势,但是没有达到显著水平(p〉0.05);在中肠中,感染嗜水气单胞菌A-OKGM组的il-1β、tnf-α、tgf-β1较感染嗜水气单胞菌对照组表达上调(p〈0.05),而il-10和il-8则相对下调。在后肠中,感染嗜水气单胞菌A-OKGM组il-1β、tnf-α、tgf-β1、il-10及il-8较感染嗜水气单胞菌对照组表达下调(p〈0.05)。本试验在日粮中添加A-OKGM1.6%对齐口裂腹鱼中肠的免疫功能有着活化的作用,对后肠中整体细胞因子起着抑制的作用,从而减轻机体产生的炎症损伤,从而增强齐口裂腹鱼肠道免疫的作用,可促进鱼体健康生长。 Abstrac t The aim of the present study was to investigate the effect of A-OKGM 1.6% added in feeds on intestinal tract biochemical indicators and cytokines gene expression profile in Schizothorax prenanti experimentally infected with Aeromonas hydrophila.After feeding C.idellus with A-OKGM and control diets for60 days,lysozyme and acid phosphatase were determined by Microplate reader;gene expression of interleukin-1β(il-1β),interleukin-10(il-10),interleukin-8(il-8),tumor necrosis factor(tnf-α) and transforming growth factor beta(tgf-β1) were determined by real time-PCR.proximal intestine(PI),middle intestine(MI) and distal intestine(DI) were collected from fish in 1 week.The results showed that compared with the injection of phosphate buffer control group,the activity of lysozyme in PI,MI,DI was increased after adding A-OKGM1.6%,but it did not reach the siglnificant level(P〈0.05),In the MI,theil-1β,tnf-α and tgf-β1 were up-regulated(P〈0.05),whileil-10 and Genomics and Applied Biology il-8 were down-regulated.,In the DI,the expression of il-1β,tnf-α,tgf-β1,il-10 and il-8 in Aeromonas hydrophila A-OKGM group were down-regulated(p约0.05).In this experiment,A-OKGM1.6% was added to the diets to regulate the immune function of the MI of the cuff,and the effect on the whole cytokines in the DI was inhibited,and the inflammatory injury caused by the body was relieved,Thereby enhancing the intestinal mucosal intestinal immune function,could promote the healthy growth of fish.
