EGCG对Tca8113细胞移植瘤小鼠肿瘤生长的抑制作用研究 被引量:2

摘要 为了探讨绿茶提取物表没食子儿茶素没食子酸酯(EGCG)对Tca8113鳞状细胞癌细胞移植裸鼠肿瘤的抑制作用及其机制。本研究选取采用Tca8113鳞状细胞癌细胞建立的裸鼠肿瘤模型48只,采用随机数字表法分为空白组(等量生理盐水灌胃)、低剂量组(ECGC 10 mg/kg灌胃)、中剂量组(ECGC 20 mg/kg灌胃)、高剂量组(ECGC 40 mg/kg灌胃)各12只,连续灌胃治疗16 d,测量各组小鼠肿瘤重量及体积、计算抑瘤率,采用流式细胞仪技术测定各组瘤体组织中细胞生长周期的分布,采用免疫组化染色技术测定各组肿瘤组织中Caspase8、Caspase3、Ki-67蛋白的表达。研究显示,低剂量组、中剂量组和高剂量组的瘤体质量和体积均显著的小于空白组(p〈0.05),且随着EGCG剂量的加大,裸鼠移植瘤体积和质量逐渐减小,而抑瘤率显著增加(p〈0.05);低剂量组、中剂量组和高剂量组的G0/G1期细胞比例显著高于空白组(p〈0.05),而S期及G2/M期细胞比例显著降低(p〈0.05);随着EGCG剂量的增加,各组G0/G1期细胞比例显著增加,而S期及G2/M期细胞比例显著降低(p〈0.05);低剂量组、中剂量组和高剂量组的肿瘤组织中Caspase8和Caspase3表达显著高于空白组(p〈0.05),而Ki-67蛋白表达显著低于空白组(p〈0.05);随着EGCG剂量的增加,各组肿瘤组织中Caspase8和Caspase3蛋白表达显著增加,而Ki-67显著降低(p〈0.05)。EGCG对Tca8113鳞状细胞癌细胞移植裸鼠肿瘤生长具有明显的抑制作用,其机制可能是通过调节细胞周期及Caspase8、Caspase3和Ki-67蛋白的表达来实现。 In order to investigate the inhibitory effect of epigallocatechin gallate(EGCG) from green tea extract on Tca8113 squamous cell carcinoma cells in nude mice and its mechanism.Select the Tca8113 squamous cell carcinoma cells to establish the tumor model in 48 nude mice,mice were randomly divided into control group(gavage with equal amount of physiological saline),low dose group(gavage with ECGC 10 mg/kg),medium dose group(gavage with ECGC 20 mg/kg) and high dose group(gavage with ECGC 40 mg/kg)(n=12),with continuous intragastric administration for 16 days.The weight and volume of tumor in each group of mice were measured and the rate of tumor inhibition was calculated.Flow cytometry was used to measure the distribution of cell growthcycle in each group of tumor tissues.Immunohistochemical staining technique was used to measure the protein expression of Caspase8,Caspase3 and Ki-67 in each group of tumor tissues.Studies have shown that the mass and volume of tumors in the low-dose group,the middle-dose group,and the high-dose group were significantly smaller than the blank group(P〈0.05).With the increase of the EGCG dose,the tumor volume and quality of the nude mice were gradually reduced,but the tumor inhibition rate was significantly increased(P〈0.05);the proportion of G0/G1 phase cells in the low-dose group,the middle-dose group,and the high-dose group was significantly higher than that in the blank group(P〈0.05),while the S phase and G2/M phase cells were significantly decreased(P〈0.05).With the increase of EGCG dose,the proportion of G0/G1 phase cells increased significantly in each group,while the proportion of cells in S phase and G2/M phase decreased significantly(P〈0.05).The expression of Caspase8 and Caspase3 in tumor tissue of low-dose group,middle-dose group and high-dose group was significantly higher than that of blank group(P〈0.05),while the expression of Ki-67 protein was significantly lower than that of blank group(P〈0.05);With increasing doses,the expression of Caspase8 and Caspase3 protein increased significantly in all groups,while Ki-67 decreased significantly(P〈0.05).EGCG significantly inhibited the growth of Tca8113 squamous cell carcinoma transplanted in nude mice.The mechanism may be achieved by regulating the cell cycle and the expression of Caspase8,Caspase3 and Ki-67 proteins.
