大黄酸对猕猴骨关节炎IL-1β、NO/iNOS表达的影响 被引量:8

摘要 [目的]观察大黄酸对猕猴骨关节炎白细胞介素-1β(IL-1β)、一氧化氮/一氧化氮合成酶(NO/i NOS)的影响,分析大黄酸对猕猴骨关节炎的治疗作用。[方法]9只健康猕猴分为正常组(A组)3只、模型对照组(B组)3只、模型治疗组(C组)3只;另外3只骨关节炎病猴纳入研究称为病猴组(D组)。正常组(A组)猕猴不行手术造模及大黄酸治疗。模型对照组(B组)和治疗组(C组)分别行前交叉韧带切断骨关节炎造模,对照组不行大黄酸治疗,而治疗组(C组)和病猴组(D组)均给予大黄酸溶液灌胃治疗。于治疗后0、2、4、8、12周时分别采集猕猴血清;双抗体夹心ELISA法检测血清样本中IL-1β和NO/i NOS浓度变化。[结果]随时间的推移,骨关节炎模型猕猴(B组)血清IL-1β、NO/i NOS的含量较正常猕猴(A组)均显著增多,两组间差异均有统计学意义(P<0.05)。使用大黄酸的模型猕猴(C组)血清IL-1β、NO/i NOS含量的增长较没有使用大黄酸的(B组)显著减缓,两组间差异有统计学意义(P<0.05)。骨关节炎病猴组(D组)血清IL-1β、NO/i NOS含量明显高于健康猴,使用黄酸后血清IL-1β、NO/i NOS逐渐降低。[结论]大黄酸抑制猕猴骨关节炎IL-1β的反应、降低i NOS的作用,减少NO的生成。 [Objective] To observe the effect of rhein on expression of interleukin 1β(IL-1β) and nitric oxide/inducible nitric oxide synthase(NO/i NOS) in rhesus osteoarthritis and to analyze the therapeutic effect of rhein on it. [Methods] Nine healthy rhesus monkeys were divided into 3 groups, 3 in the normal group(group A), 3 in the control group(group B), and 3 in the treatment group(group C). In addition, 3 monkeys with osteoarthritis were also used in the study termed as the osteoarthritic monkey group(group D). The normal group(group A) was not treated with surgery either rhein. The control group(group B) and treatment group(group C) received cutting anterior cruciate ligament to creat osteoarthritis models. After that, the control group was not treated with rhein, while the treatment group(group C) and osteoarthritic monkeys group(group D) were treated with rhein by intragastric administration in solution. The rhesus serums were collected at 0, 2, 4, 8 and 12 weeks after treatment, and the level of IL-1β and NO/i NOS in the serum were detected by sandwich ELISA with double-antibodies. [Results] Compared with the normal animals(the group A), the serum IL-1β and NO/i NOS in the osteoarthritic models(the group B) gradually in-creased over time with significant differences between them(P〈0.05). However, the levels of IL-1β and NO/i NOS in the group C treated with rhein rose remarkably slower than the group B with significant differences between them(P〈0.05). In addition, the Serum levels of IL-1β and NO/i NOS in the osteoarthritic monkey group(the group D) were significantly higher, which decreased gradually after using rhein.[Conclusion] The Rhein inhibits the IL-1β response in rhesus osteoarthritis, decreases the effect of i NOS and reduces the production of NO.
