摘要
目的研究耳鸣发生过程中内膝体不同亚核团内抑制性系统的功能变化。方法用慢性水杨酸制剂法制作小鼠耳鸣模型后,制备内膝体急性脑片,采用全细胞膜片钳技术分别在脑片上内膝体的腹侧核和背侧核记录神经元的自发抑制性突触后电流(sIPSC)。结果模型组的内膝体腹侧核神经元上sIPSC的频率明显降低(P<.0.01),sIPSC的幅度、上升和衰减时间与对照组相比无明显差异(P>0.05)。对照组和模型组内膝体背侧核神经元上sIPSC的频率、幅度、上升和衰减时间均无明显差异(P>0.05)。结论水杨酸钠诱导耳鸣的过程中可以选择性地损伤内膝体腹侧核中GABAA受体介导的自发突触反应。
Objective This study aimed to explore the changes in the function of inhibitory system within different subnuclei of the medial geniculate body(MGB)during tinnitus.Methods The innitus mice model was induced by chronic administrating of sodium salicylate,after that,acute brain slices containing MGB were prepared and then whole-cell patch clamp technique was applied for recording the spontaneous inhibitory postsynaptic currents(sIPSCs) on the neurons in the ventral MGB(MGV)and dorsal MGB(MGD)respectively.Results For the neurons in the MGV,the frequency of sIPSCs was significantly reduced in tinnitus model group(P.0.01),there is no difference in the amplitude,rising and decay time of the sIPSCs between model and control groups(P〈0.05).For the neurons in the MGD,there is no difference in the frequency,amplitude,rising and decay time of the sIPSCs between model and control groups(P〈0.05).Conclusion The GABAA receptor-induced spontaneous synaptic responses in the MGV can be selectively impaired during sodium salicylate-induced tinnitus.
