科罗索酸通过抑制Hippo-YAP信号转导通路促进非小细胞肺癌细胞的凋亡 被引量:7

摘要 目的:研究科罗索酸对肺癌细胞增殖与凋亡的影响及其机制。方法:MTT、Caspase3/7活性检测、Western blot等方法检测不同浓度的科罗索酸是否影响肺癌细胞株A549的增殖和凋亡;Western blot、免疫荧光实验等方法检测科罗索酸对肺癌细胞A549 Hippo通路中YAP蛋白进行定量和定位的检测。结果:MTT结果显示科罗索酸能够抑制肺癌细胞株A549的增殖,半抑制浓度为40μmol/L;Caspase活性检测结果显示科罗索酸能够促进肺癌细胞株A549的凋亡,半致死浓度为40μmol/L;Western blot、免疫荧光试验结果显示科罗索酸能明显抑制肺癌细胞株A549中YAP蛋白的表达。结论:科罗索酸可能通过调控Hippo-YAP信号通路抑制肺癌细胞的增殖并促进其凋亡。 Objective:To explore the possible mechanism of Corosoic acid in the apoptosis and proliferation of lung cancer cells. Methods:MTT assay,Caspase 3/7 activities and Western blot were used to determine the effect of Corosolic acid on the proliferation and apoptosis of non-small cell lung cancer cells;Western blot and immunoflurensence were used to detect the expression and location of YAP after treatment hepatoma cell line with CA. Results:MTT test showed that Corosoric acid could inhibit the proliferation of A549,and the IC50 value was 40 μmol/L. Caspase activity test showed that Corosolic acid could promote apoptosis of lung cancer cell line A549 which was consistent with the level of Caspase substrates by Western blot. Western blot and immunofluorescence test showed that corosolic acid could significantly inhibit the expression of YAP protein in lung cancer cell line A549. Conclusion:Corosolic acid can inhibit the proliferation and apoptosis of lung cancer cells by regulating Hippo-YAP signaling pathway.
