Cloning and Expression Analysis of MeSnRK2-1 Gene in Cassava

摘要 蔗糖非发酵型蛋白激酶SnRKs在植物应对非生物胁迫的过程中具有非常重要的功能。本研究利用PCR技术从木薯栽培种KU50中克隆得到一个SnRK2家族基因,命名为MeSnRK2-1,对其进行了序列比对和进化树分析,并利用荧光定量技术对该基因在多种处理下的表达模式进行分析。结果表明,该基因的开放阅读框全长为1 062 bp,编码354个氨基酸,进化树分析显示MeSnRK2-1与橡胶树和麻疯树中的同源蛋白亲缘关系较近。此外,荧光定量结果表明MeSnRK2-1在转录水平受到高盐胁迫、H2O2和低温的诱导作用的同时也受到了ABA和甘露醇的抑制作用。这些结果表明MeSnRK2-1在多种非生物胁迫下的信号转导通路中可能具有重要的功能。本研究为后续深入探索MeSnRK2-1的功能以及木薯的逆境生理提供理论和物质基础。 Sucrose non-fermenting 1-related protein kinases (SnRKs) play an important role in the response of plants to adversity stress. In the present study, we cloned a SnRK2 family gene named MeSnRK2-1 from the cassava cultivar KU50 by PCR method. Sequences alignment and phylogenetic analysis were carried out and the expression patterns of MeSnRK2-1 under various treatments were investigated by florescent quantitation. The results showed that MeSnRK2-1 had a 1062 bp open reading frame and encoded 354 amino acids. Phylogenetic analysis showed that MeSnRK2-1 was closely related to homologous proteins in Hevea brasiliensis and Jatropha curcas. In addition, florescent quantitation result revealed that MeSnRK2-1 was induced by salt stress, H2O2 and low temperature, but inhibited by ABA and mannitol at the transcriptional level. These results suggested that MeSnRK2-1 might play an important role in signal transduction pathways under various abiotic stresses. This study could provide the theoretical and material foundation for further exploration of the function of MeSnRK2-1 and stress physiology in cassava.
