香牙蕉与枯萎病4号小种抗性相关的SCAR分子标记开发 被引量:6

Development of SCAR Molecular Marker Related with Resistance to Fusarium oxysporum f. sp. Cubense Race 4 in Banana(Musa AAA Cavendish)

摘要 香蕉枯萎病是香蕉的一种毁灭性病害,采用抗病品种是控制病害的有效途径。目前香蕉抗病或耐病品种选育效率低,需要长期的田间观察和表征。分子标记辅助选择可以大大的提高传统育种的效率和准确性。香牙蕉是香蕉贸易中的主要类型,也是中国主栽的香蕉类型。本研究从100对SRAP分子标记中筛选出一对引物me9-em5,其在感病香牙蕉扩增出一条约400 bp的特异条带,抗病香牙蕉没有该条带的扩增。通过回收测序,设计并获得引物SC11-SC12,将SRAP分子标记成功转化为可以鉴定香牙蕉对枯萎病的抗感病性的SCAR标记。该标记可以鉴定本研究中涉及的香牙蕉的枯萎病抗感病性,且准确率达到96.875%。在香蕉抗枯萎病育种中具有较高的应用价值。 Fusarium wilt is one of the most devastating disease of banana (Musa spp.L.). The use of resistant cultivars is an effective method for the disease control. At present, the efficiency of resistant or tolerant banana cultivars development is very low because the results must be confirmed by many years of field observation and evaluation, while molecular marker-assisted selection can greatly improve the efficiency and accuracy of traditional breeding. Musa AAA Cavendish which is the main types of banana trade, is also the main type of banana cultivars in China. In this study, we screened a pair of primers me9-em5 out from 100 pairs of SRAP molecular markers, from which we amplified a 400 bp specific band in the susceptible Musa spp., but no amplification of the band in the resistant Musa spp. By recycling and sequencing, the study designed and obtained primer SC11-SC12, and successfully transformed SRAP molecular marker into the SCAR marker which could authenticate the disease resistance of Musa spp. to Fusarium wilt. The marker could identify the disease resistance to Fusarium wilt in this study and the precision rate was 96.875%, which had a high application value in in the breeding of Fusarium wilt resistance of banana.
