FTIR结合DNA条形码鉴定黄花蒿及其近缘植物 被引量:1

Combination of FTIR and DNA Barcode for Identification of Artemisia annual L. and Its Closely Related Species

摘要 黄花蒿是治疗痢疾的特效中药,近缘种与黄花蒿混杂出售严重影响了黄花蒿质量。本研究采用傅里叶红外光谱法(FTIR)和DNA条形码对黄花蒿及近缘种进行了鉴定。FTIR分析结果表明,黄花蒿及其近缘种中芳香类、酰胺类及萜类化合物种类多、含量丰富。黄花蒿及其近缘种的二阶导数图谱可将黄花蒿与其混淆种区分开。红外光谱进行系统聚类分析后,6个物种可分为两组,黄花蒿为一组,其余5个种聚为一组。候选的4条序列(ITS,ITS2,rbc L,psb K-psb I)中,ITS2序列具有较好的PCR扩增成功率和测序成功率,在所考察的候选序列中种间变异和种内变异较高,种间变异大于种内变异,两者形成了较为明显的Barcoding gap区。FTIR辅以ITS2序列将可精准地鉴别中药材黄花蒿。 A rtemisia annual L. is a special traditional Chinese medicine for the treatment of dysentery, and the mixed sale of closely related species and Artemisia annua has seriously affected the quality of A. annual. In this paper, fourier transform infrared spectroscopy (FTIR) and DNA barcoding sequences were used to identify A. annual and its closely related species. FTIR analysis showed that the aromatic, amide and terpenoids were rich in species and abundance in A. annual and its closely related species. The second derivative spectrum of A. annual and its closely related species could seperate them apart. Based on the cluster analysis of infrared spectra, six species could be divided into two groups, with A. annual in a group, and the other five species in another group. Among the four candicate sequences (ITS, ITS2, rbcL, psbK-psbI), ITS2 had good PCR amplification success rate and sequencing success rate. Interspecific variation and intraspecific variation were higher in the four sequences, and interspecific variation was greater than intraspecific variation, and the two formed a more obvious barcoding gap region. FTIR combined with ITS2 sequence could be able to accurately identify A. annual.
