玉米赤霉烯酮对小鼠T淋巴细胞共刺激分子调节功能的影响 被引量:3

Effect of zearalenone on the modulation of mouse T lymphocyte costimulatory molecules in vitro

摘要 在原代培养的小鼠T淋巴细胞中加入刀豆蛋白A(Con A)作为活化刺激剂,研究不同浓度玉米赤霉烯酮(ZEA)对T淋巴细胞共刺激信号分子CD28和CD152,及PI3K-Akt-mTOR信号通路关键蛋白表达的影响。结果表明:染毒48h后,Con A组与细胞空白组相比,CD28和CD152均显著升高;染毒组与Con A组相比,10和20μmol·L-1ZEA组CD28表达量呈下降趋势但差异不显著,CD152表达量显著或极显著降低,40μmol·L-1 ZEA组CD28表达量显著降低,CD152表达量无显著差异。染毒24h后,Con A组与细胞空白组相比,PI3K等蛋白表达量显著或极显著升高;各染毒组与Con A组相比,PI3K等蛋白表达量均显著或极显著下降,且呈剂量依赖性。这一研究说明ZEA可通过干扰CD28/CD152表达及抑制PI3K-Akt-mTOR信号通路来干扰T淋巴细胞的活化过程。 The concanavalin A (Con A) was added to primary cultured mouse T lymphocyte cells as an activation stimulator to study the expression of CD28 and CD152 co-stimulation signal molecules of T lymphocytes with different concen- trations of zearalenone (ZEA). In addition, the effect of key protein expression in PI3K-Akt-mTOR signaling pathway was investigated. The results showed that after 48 hours of exposure, compared with the blank cells group, the expression of CD28 and CD152 in the Con A group was significantly increased. Compared with the Con A group, the expression of CD28 in 10 and 20 /zmol·L^-1 ZEA group showed a decreasing trend with no significant difference. The expression of CD152 was significantly or very significantly decreased. The expression of CD28 in 40 btmol·L^-1 ZEA group was significantly decreased, while the expression of CD152 was not significantly different. After 24 hours of exposure, compared with the blank cells group, the protein expressions of PI3K, Akt and mTOR were significantly or very significantly increased in the Con A group. Compared with Con A group, PI3K, Akt, roTOR and other proteins were significantly or very significantly decreased in the exposure groups, and in a dose-dependent manner. ZEA can interfere with the activation of T lymphocytes by interfering with the expression of CD28/CD152 and inhibiting the PI3K-Akt-mTOR signaling pathway.
