摘要
目的探讨酸枣仁提取物通过调节慢波睡眠从而促进动物增高的作用及其作用机制。方法给予小鼠酸枣仁提取物低剂量和高剂量(0.18 mg/g,0.32 mg/g体重灌胃),与空白对照组比较,观察酸枣仁提取物是否促进发育期小鼠体长的增长,同时给予大鼠酸枣仁提取物,观察脑组织5-羟色胺1A受体(5-HT1AR)表达,慢波睡眠的变化,体内生长激素含量变化,阐明酸枣仁提取物促进骨骼增长的作用是通过增加5-HT1AR表达而延长慢波睡眠进而促进生长激素分泌实现的。结果酸枣仁提取物可以促进发育期小鼠体长增长,灌胃10 d和20 d后高、低剂量组体长较空白对照组明显增长(P<0.01);高剂量具有更好的增高效果;同时高剂量组大鼠5-HT1AR活性较空白对照组增高(P<0.01),慢波睡眠较空白对照组有所延长(P<0.01),而且血清生长激素含量较空白对照组升高(P<0.01);而5-HT1AR选择性抑制剂可以抑制这一现象。结论酸枣仁提取物可以促进动物体长的增长,其作用机制可能是通过提高脑组织5-HT1AR含量而与5-HT结合,延长慢波睡眠时长,从而促进生长激素的分泌引起的。
Objective To study the extraction of semen ziziphispinosae( zs) by adjusting the slow wave sleep so as to promote the growth of animal body length and its mechanism of action. Methods The mice were given extraction of semen ziziphispinosae( zs) with low dose and high dose( 0. 18,0. 32 mg/g weight for intragastric administration). Compared with blank control group,the body length of pubertal mice was observed. The changes of growth hormone content,5-hydroxytryp 1-A receptor content changes and the slow wave sleep changes were observed to clarify the role of the extraction of semen ziziphispinosae( zs) which promotes the bone growth by the extension of the slow wave sleep and improving the growth hormone secretion. Results The extracts of semen ziziphispinosae could promote the growth of mice in the developmental stage. After 10 days and 20 days of intragastric administration,the body length of the high and low dose groups increased significantly( P〈0. 01); High doses had a better effect. At the same time,the activity of 5-HT1 AR in high dose group was higher than that in blank control group( P〈0. 01) and slow wave sleep was longer than the blank control group( P〈0. 01). The serum growth hormone content was higher than that of the blank control group( P〈0. 01). 5-HT1 AR selective inhibitors can inhibit this phenomenon. Conclusion The extract of semen ziziphispinosae( zs) can promote the growth of animal body length,and its mechanism maybe extend the slow wave sleep time,thereby promote the secretion of growth hormone by increasing the brain 5-HT1 AR content which combined with 5-HT.
