摘要
目的研究颈椎病治疗仪对颈椎病模型大鼠颈椎骨组织中Ca、P及血清中IL-lβ、IL-6、TNF-α含量的影响,探讨颈椎病治疗仪防治颈椎病的作用机制。方法 2月龄SD大鼠48只,适应性喂养1周,随机分为空白组12只和造模组36只。造模组予以切除C2~C7棘上韧带、棘间韧带和分离椎旁两侧肌肉,建立颈椎退变动物模型;造模3个月后,抽签法随机分为3组,即模型组、治疗组和对照组各12只。空白组和模型组不予任何治疗;治疗组和对照组分别给予颈椎病治疗仪和低频电子脉冲治疗仪治疗,每次30 min,每日1次。治疗4周后,处死动物,获取各组大鼠的C5、C6骨组织和血清,用半自动生化分析仪测定各组C5、C6骨组织中Ca、P含量和放射免疫法测定血清中IL-lβ、IL-6、TNF-a含量。结果与空白组比较,模型组中Ca、P的含量下降,IL-1β、IL-6、TNF-α含量增加(P均<0.01);治疗组和对照组治疗后,Ca、P含量均升高,IL-1β、IL-6、TNF-α含量均降低,与模型组比较,差异均有统计学意义(P<0.05);但治疗组与对照组比较,差异无统计学意义(P>0.05)。结论颈椎病治疗仪可通过提高颈椎骨组织中Ca、P含量,下调血清中IL-1β、IL-6和TNF-α炎性细胞因子含量,抑制颈椎退变。
Objective: To investigate the effect of cervical spondylosis treatment apparatus(CSTA) on calcium(Ca) and phosphorus(P) in degenerated cervical bone and IL-lβ, I L-6 and TNF-a in serum in rats with cervical spondylopathy(CS), so as to explore mechanisms of CSTA in the prevention and treatment of CS. Methods: After one week of acclimation, two-month-old male Sprague Dawley rats(n=48) were randomly divided into blank group(n=12) and model group(n=36). The models were established via cutting off C2-C7 supraspinal and interspinal ligaments and seperated the muscle groups beside cervical vertebra. Three monthes later, the animals were randomly divided into three groups, including model group(n=12), treatment group(n=12) and control group(n=12). The blank group and model group did not receive any treatment. The treatment group and control group were treated with the CSTA and low frequency electronic pulse therapy apparatus respectively, daily, 30 min each time. After four weeks treatment, the rats were sacrificed to collect C5, C6 cervical bone and serum. Automatic biochemistry analyzer was used to detect the content of Ca and P in every group, and radioimmunity assay was applied to measure the content of IL-lβ, IL-6 and TNF-a in serum. Results: Compared with the blank group, Ca and P levels in model group were significantly decreased( P〈0.01), and IL-1β 、IL-6 、TNF-α were significantly increased(P〈0.01). Compared with the model group, the content of Ca and P were increased, and IL-1β, IL-6 and TNF-αwere decreased after treatment in the treatment group and the control group(P〉0.05). However, there was no statistical difference between the treatment group and the control group(P〈0.05). Conclusion: The cervical spondylosis treatment apparatus could inhibit the degeneration of the cervical vertebra by increasing the content of Ca and P in cervical vertebra and decreasing IL-1β, IL-6 and TNF-α in serum.
