CSE1L在绝经后女性骨关节炎滑膜细胞中的作用探究

A Study with Regard to How CSE1L Works in Osteoarthritis among Postmenopausal Women

目的:探讨CSE1L在绝经后女性骨关节炎滑膜细胞中的表达及其作用和机制。方法:通过RT-PCR、Western blot以及免疫组化的实验方法检测和比较CSE1L在绝经后女性骨关节炎和创伤截肢患者关节滑膜细胞中的表达,不同浓度TNF-α或TNF-α抗体处理的关节炎滑膜细胞中CSE1L的表达;采用si RNA干预其骨关节炎滑膜细胞中CSE1L的表达后,通过RT-PCR、ELISA以及Western blot检测其TNF-α的表达和分泌情况。结果:RT-PCR以及Western blot结果均提示绝经后女性骨关节炎滑膜细胞中CSE1L的表达较创伤截肢患者关节滑膜细胞显著增高(P<0.01,P<0.001)。对CSE1L进行si RNA干预降低其表达水平后,CSE1L表现出与TNF-α浓度呈正相关的变化趋势;此外,在对CSE1L进行敲减以降低其表达水平后,绝经后女性骨关节炎滑膜细胞中TNF-α的表达水平变化提示其受到CSE1L调节。结论:CSE1L可能通过调节TNF-α的表达促进绝经后女性骨关节炎的发展。

Objective： To investigate whether the expression of CSE1 L elevates in the synoviocytes among postmenopausal women with osteoarthritis（OA）,after the positive result came out,to futher preliminary look into the mechanism of how CSE1 L works in the synoviocytes among postmenopausal women with OA.Methods： Investigate whether the expression of CSE1 L elevates in the synoviocytes among postmenopausal women with OA by means of RT-PCR,western blot and immunohistochemistry,and then conduct quantitative and semi-quantitative analysis.After knocking down the expression of CSE1 L with CSE1 L specific si RNA,to investigate whether CSE1 L shows a positive relationship with the severity of OA or not by means of western blot.Through the methods of RT-PCR,ELISA and western blot,the interaction between CSE1 L and TNF-α was preliminarily illustrated,so the mechanism of how CSE1 L works in the synoviocytes among postmenopausal women with OA was preliminarily showed up.Results： In the synociocytes among postmenopausal women with OA,the results of both RT-PCR and western blot manifested that the expression of CSE1 L elevated significantly（P〈0.01,P〈0.001）.Moreover,after knocking down the expression of CSE1 L with CSE1 L specific si RNA,the expression level of CSE1 L show a positive relationship with TNF-α concentration.In addition,after knocking down the expression of CSE1 L with CSE1 L specific si RNA,and the expression level of TNF-α came up to be regulated by CSE1 L in the synoviocytes among postmenopausal women with OA.Conclusions： CSE1 L promote OA development via interaction with TNF-α expressed by the synoviocytes among postmenopausal women with OA.
