蝙蝠葛苏林碱对大鼠离体脑片氧糖剥夺损伤的保护作用 被引量:3

摘要 目的:探讨蝙蝠葛苏林碱(daurisoline,DS)对大鼠脑缺血所致神经细胞损伤的保护作用及作用机制。方法:采用离体大鼠脑片培养,通过氧糖剥夺(oxygen-glucose deprivation,OGD)模拟载体脑缺血损伤模型。脑片经TTC染色,用酶标仪测定吸光度值和乳酸脱氢酶(lactate dehydrogenase,LDH)释放量。结果:TTC染色显示,蝙蝠葛苏林碱各剂量组脑片组织损伤百分率分别下降至(51.37±0.48)%、(33.86±0.27)%、(25.09±0.75)%;LDH释放率分别下降至(53.41±3.89)%、(38.03±3.71)%、(28.03±3.71)%;与OGD组比较,差异有显著统计学意义(P<0.01)。结论:蝙蝠葛苏林碱可以减轻OGD引起的脑片损伤,并呈剂量依赖性。 Objective:To explore the neuroprotective effect of Daurisoline on brain ischemia and mechanism.Methods:The cultured rats' brain slices exposed to oxygen-glucose deprivation( OGD) injury.Brain slices injury was evaluated by MTT staining and the release of lactate dehydrogenase( LDH) was determined.Results:The TTC staining shown that the injury percentage of rats' brain slices with Daurisoline cultured was decreased to( 51.37 ± 0.48) %,( 33.86 ± 0.27) % and( 25.09 ± 0.75) %; The percentage of LDH releasing was decreased to( 53.41 ± 3.89) %,( 38.03 ± 3.71) % and( 28.03 ± 3.71) %,respectively.There were statistical significance to compare with the OGD model group( P〈0.01).Conclusion:Daurisoline can exert neuroprotection against OGD-induced injury in cultured brain slices of rats.
