摘要
目的:探究复方钩藤降压片对SHR左室肥厚及血清瘦素和脂联素的影响。方法:将32只SHR雄性大鼠随机分为模型组、复方钩藤降压片组、替米沙坦组,每组分别为8、16、8只。另外,选取8只雄性WKY大鼠作空白对照组;各组均予自由饮食、饮水。复方钩藤降压片组灌胃给予43.2 mg/m L复方钩藤降压片,替米沙坦组灌胃给予0.36 mg/m L替米沙坦,模型组和空白对照组灌胃给予等体积的蒸馏水,共8周;取左室小块心肌,观察HE染色后心肌组织的形态学改变;腹主动脉取血,用酶联免疫吸附法测定血清中LP及APN表达情况。结果:模型组心肌细胞分布紊乱,细胞直径增大,细胞核深染明显增大,心肌纤维明显增粗,视野下有断裂现象,复方钩藤降压片组心肌细胞轻度肥大,细胞间质轻度增生,心肌纤维无明显增粗,心肌纤维较模型组有减少,无断裂现象;模型组血清LP水平明显高于空白对照组(P<0.01),复方钩藤降压片组、替米沙坦组血清LP水平低于模型组(P<0.01);模型组血清APN水平明显低于空白对照组(P<0.01),复方钩藤降压片组、替米沙坦组血清APN水平高于模型组(P<0.01)。结论:复方钩藤降压片可延缓SHR左室重构(LVRM),减慢心肌纤维化进程,其作用机制可能与降低血清LP和升高血清APN含量有关。
Objective: To observe the effect of Fufang Gouteng Jiangya tablet on SHR' left ventrieular hy- pertrophy, serum leptin (LP) and adiponectin (APN). Methods: A total of 32 male SHR were randomly divided into model group (8 rats), Fufang Gouteng Jiangya tablet group (16 rats) and telmisartan group (8 rats). The other 8 male rats served as the control group. Groups were fed on free diet and drinking water. The Fufang Gouteng Jiangya tablet group took 43.2 mg/mL solution by intragastric administration at 10 mL/kg every day. By contrast, tehnisartan group took the mixture of 0.36 mg/mL solution in the same way while the model group and the con- trol group took distilled water. This experiment lasted for 8 weeks. The left ventricular myocardium was taken to observe the morphological changes of cardiac tissue after HE staining. Blood was taken from the abdominal aor- ta and the expression level of LP and APN in serum was determined by ELISA assay. Results: The model group rats myocardial cells were distributed in disorder, cell diameter increased, the nuclei were hyperchromatic and obviously increased, myocardial fibers enlarged and a fracture phenomenon was seen under the field of vi-sion. The Fufang Gouteng Jiangya tablet group rats myocardial cells were of mild hypertrophy, intercellular sub- stance of mild hyperplasia and no obvious enlargement of myocardial fibers. Compared with model group, my- ocardial fibers decreased and there was no fracture phenomenon. The serum LP level in model group was obvi- ously higher than that of the control group(P〈0.01 ). The serum LP level in Fufang Gouteng Jiangya tablet group and telmisartan group is lower than that of the model group (P〈0.01); The serum APN level in model group was obviously lower than that of the control group (P〈0.01). The serum APN level in Fufang Gouteng Jiangya tablet group and telmisartan group is higher than that of the model group (P〈0.01). Conclusion: Fufang Gouteng Jiangya tablet delays hypertensive left ventricular remodeling (LVRM) and slows down the process of myocardial fibrosis. The mechanism may be related to the decrease of serum LP and the increase of serum APN level.
