摘要
目的探讨血清人微小RNA-17-5p(hsa-miR-17-5p)和hsa-miR-19b-3p的表达与儿童哮喘的关系。方法采用实时荧光定量检测80例哮喘儿童(哮喘组)和60例健康儿童(对照组)血清hsa-miR-17-5p和hsa-miR-19b-3p的相对表达水平,采用ROC曲线分析两者诊断哮喘的意义。结果哮喘组血清hsa-miR-17-5p表达水平高于对照组,hsa-miR-19b-3p表达水平低于对照组(P<0.05);hsa-miR-17-5p和hsa-miR-19b-3p联合诊断哮喘的ROC曲线下面积(0.815)优于hsa-miR-17-5p(0.787)或hsa-miR-19b-3p(0.608);生物信息学分析显示,hsa-miR-17-5p和hsa-miR-19b-3p的靶基因编码蛋白主要参与炎症、免疫和转录效率等。结论 Hsa-miR-17-5p在哮喘儿童血清中高表达,hsa-miR-19b-3p低表达,有可能作为诊断哮喘的标志物;其机制可能与炎症、免疫和转录效率有关。
Objective To explore the relationship between serum hsa-miR-17-5p and hsa-miR-19b-3p expressions and asthma in children.Methods The relative expression levels of serum hsa-miR-17-5p and hsa-miR-19b-3p were determined by fluorescence quantitative PCR in 80 children with asthma(group A)and 60 healthy children(group B).The receiver operating characteristic(ROC)curve was drawn to analyze the significance of the target genes hsa-miR-17-5p and hsa-miR-19b-3p in the diagnosis of asthma.Results Serum expression level of hsa-miR-17-5p was significantly higher,but that of hsa-miR-19b-3p was lower,in group A than those in group B(P〈0.05).When combined detection of hsa-miR-17-5p and hsa-miR-19b-3p in the diagnosis of asthma,the area under ROC curve was 0.815,which was higher than 0.787 for hsa-miR-17-5p or 0.608 for hsa-miR-19b-3p alone.Bioinformatics analysis showed that the proteins encoded by the target genes of hsa-miR-17-5p and hsa-miR-19b-3p were mainly involved in the inflammation,immunity and transcriptional efficiency.Conclusion Hsa-miR-17-5p is highly expressed and hsa-miR-19b-3p is lowly expressed in the serum of asthmatic children,which may become the markers for the diagnosis of asthma.The mechanism for that may be related to the inflammation,immunity and transcription efficiency.
