摘要
目的:采用MTT法评估比较几种常见的市售人工泪液对人角膜上皮细胞(HCECs)的毒性。方法:常规培养HCECs,用完全培养基DMEM将几种人工泪液分别稀释2倍、4倍和10倍,MTT法检测不同浓度滴眼液处理细胞24 h后的细胞存活率。结果:滴眼液稀释2倍时,所有滴眼液都有显著性细胞毒性,但其余组和小牛血去蛋白提取物滴眼液组相比,细胞毒性更显著(P<0.01),稀释4倍时,除了小牛血去蛋白提取物滴眼液组和右旋糖酐羟丙甲纤维素滴眼液组,其余组均表现出显著的细胞毒性(P<0.001),稀释10倍时,小牛血去蛋白提取物滴眼液组、玻璃酸钠滴眼液1组和右旋糖酐羟丙甲纤维素滴眼液组没有显著的细胞毒性(P>0.05)。结论:小牛血去蛋白提取物滴眼液、对HCECs毒性最小。
To evaluate the cytotoxicity of several common commercial artificial tears to human corneal epithelial cells(HCECs) in vitro,HCECs were cultured in different drugs with the final concentration of 50%, 25%, 10%.Cell proliferation was evaluated by MTT assay. The results showed that, the cytotoxicity of artificial tears to HCECs was concentration-dependent. All kinds of eye drops showed significant cytotoxicity when they were diluted 2 times. Compared with deproteinized calf blood extract eye drops, the others showed greater cytotoxicity(P 〈 0.01). When they were diluted 4 times, except for the deproteinized calf blood extract eye drops and dextran and hypromellose eye drops group,the others showed significant cytotoxicity(P 〈 0.001). When they were diluted 10 times,the deproteinized calf blood extract eye drops, sodium hyaluronate eye drops 1 and dextran and hypromellose eye drops had no cytotoxicity(P 〉 0.05). The deproteinized calf blood extract eye drops presented the lower toxicity than the other commercial available artificial tears.
