摘要
目的探讨热射病模型大鼠脾脏巨噬细胞的极化方向。方法取清洁级Wistar大鼠40只(36只+储备4只),适应性生长2周后开始用于实验。将实验动物分为热暴露即刻组、热暴露后1 d、2 d、3 d、7 d组、对照组共6组,每组6只。热射病造模成功后提取各组大鼠脾脏巨噬细胞,用细胞化学免疫法鉴定脾脏巨噬细胞。通过实时定量PCR、Western blotting法检测各组诱导型一氧化碳合酶(i NOS)、Ⅰ型精氨酸酶(ArgⅠ)的表达。结果热处理后,各热暴露组与对照组相比,i NOS mRNA及蛋白除热暴露后7 d组差异无统计学意义(P>0.05),余各组均降低(P均<0.05);热暴露后1 d组ArgⅠmRNA及蛋白降低(P均<0.05),热暴露后2 d、3 d、7 d组逐渐升高(P均<0.05)。结论热射病模型大鼠脾脏巨噬细胞向M2型方向转化,未向M1型方向转化。
Objective To investigate the polarization direction of the splenic macrophages of heat-stroke rat models.Methods Forty clean Wistar rats(36 for experiment and 4 for substitute) were selected in the experiment after 2-week adaptive growth. The rats were randomly divided into the following groups: heat exposure immediate group,1-day,2-day,3-day,7-day thermal exposure groups,and control group with 6 in each. After the successful modeling,the splenic macrophages of rats were extracted and identified by the way of cytochemical immunization. The real-time quantitative PCR and Western blotting were used to test the expression of nitric-oxide synthase(i NOS) and Arg Ⅰ. Results The experiment showed that there was a significant difference in the i NOS among all groups except the 7-day thermal exposure group,and the i NOS decreased in the five other groups(all P 〈 0. 05). The level of Arg Ⅰ of the 1-day thermal exposure group gradually increased(P 〈 0. 05). Conclusion The splenic macrophages of the heat-stroke rat models showed a conversion to Type M2 instead of Type M1.
