丁苯酞预处理对急性低压低氧小鼠记忆功能及海马乙酰胆碱含量的影响 被引量:1

Effects of Butylphthalide Preconditioning on the Memory Function and Hippocampal Acetylcholine Content in Mice with Acute Hypobaric Hypoxia

摘要 目的观察丁苯酞(NBP)预处理对急性低压低氧小鼠记忆功能及海马乙酰胆碱(Ach)含量的影响。方法将42只成年昆明小鼠随机分为3组,即常压常氧对照组、低压低氧对照组和低压低氧实验组。通过Morris水迷宫实验的逃跑潜伏时间和平均速度评估小鼠空间记忆功能,检测海马组织中Ach含量及乙酰胆碱转移酶(ChAT)活性。结果与常压常氧对照组比较,低压低氧对照组和低压低氧实验组潜伏期明显延长,分别为(26.39±8.44)s、(44.60±7.80)s、(35.97±4.49)s,平均速度显著减慢,分别为(24.91±4.61)cm/s、(7.69±4.33)cm/s、(13.58±5.33)cm/s;海马Ach含量、ChAT活性显著降低(P<0.05或P<0.01),Ach含量分别为(8.11±0.35)mg/g、(7.56±0.42)mg/g、(8.00±0.28)mg/g,ChAT活性分别为(271.0±70.1)U/g、(167.4±46.2)U/g、(238.4±65.4)U/g;与低压低氧对照组比较,低压低氧实验组的潜伏期显著缩短,平均速度显著增快,海马Ach含量、ChAT活性显著升高(P<0.05或P<0.01)。结论 NBP预处理可以减轻急性低压低氧对小鼠记忆功能的损害,可能与Ach含量及ChAT活性增加有关。 Objective To observe the effects of butylphthalide( dl-3-n-butylphthalide,NBP) preconditioning on the level of acetylcholine( Ach) in hippocampus and memory in mice with acute hypobaric hypoxia.Methods Forty-two Kunming mice were randomly divided into normobaric normoxia control group,hypobaric hypoxia control group and hypobaric hypoxia experimental group.The spatial memory was estimated by escape latence and average velocity of Morris water maze test.Meanwhile,the content of Ach and the activity of choline acetyltransferase( ChAT) were measured.Results Compared with the normobaric normoxic control group,the latency of the hypobaric hypoxia control group and the hypobaric hypoxia test group was significantly prolonged,which were( 26.39±8.44) s,( 44.60±7.80) s,( 35.97±4.49) s,respectively,and the average velocity was significantly slowed down,which were( 24.91±4.61) cm/s,( 7.69±4.33) cm/s,( 13.58±5.33) cm/s respectively.The Ach content and ChAT activity in hippocampus were significantly decreased( P〈0.05 or P〈0.01),and the Ach contents were( 8.11±0.35) mg/g,( 7.56±0.42) mg/g,( 8.00±0.28) mg/g,respectively.The ChAT activities were( 271.0±70.1) U/g,( 167.4±46.2) U/g,( 238.4±65.4) U/g,respectively.Compared with the hypobaric hypoxia control group,the incubation period of the hypobaric hypoxia test group was significantly shortened,the average speed was significantly increased,and the hippocampal Ach content and ChAT activity were significantly increased( P〈0.05).Conclusion NBP preconditioning can alleviate the damage of acute hypobaric hypoxia on memory function in mice,which may be related to the increase of Ach content and ChAT activity.
