转化生长因子-β1基因多态性与新疆地区维汉妊娠期糖尿病易感性的关系分析 被引量:4

Relationship between gene polymorphism of transforming growth factor-β1 and susceptibility to gestational diabetes mellitus in Uygur and Han nationalities of Xinjiang Autonomous Region

摘要 目的分析转化生长因子-β1(TGF-β1)基因多态性与新疆地区维汉妊娠期糖尿病(GDM)易感性的关系。方法选取2014年6月至2017年12月新疆乌鲁木齐市妇幼保健院收治的孕妇450例,依据GDM诊断标准分为GDM组(确诊为GDM,200例,其中维、汉族各100例)和对照组(正常孕妇,250例,其中维、汉族分别为100例、150例),比较两组TGF-β1水平,采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)技术对TGF-β1基因单核苷酸多态性(SNP):-509C/T、+869T/C、+915G/C进行测定分析。结果 GDM组血清TGF-β1水平高于对照组,且GDM组中维吾尔族患者TGF-β1水平高于汉族患者(P<0.05);两组-509C/T基因位点的CC、CT、TT基因型频率及+915G/C位点的GG、GC、CC基因型频率比较差异无统计学意义(P>0.05),GDM组、对照组+869T/C基因位点的TT、TC、CC基因型频率差异有统计学意义(P<0.01);GDM组+869T/C等位基因T、C频率与对照组比较差异有统计学意义(P<0.01);GDM组中维吾尔族患者+869T/C基因位点的TT、TC、CC基因型频率、等位基因(T、C)频率与汉族患者比较差异有统计学意义(P<0.01)。结论 TGF-β1基因位点+869T/C单核苷酸多态性可能在新疆地区维汉GDM易感性中起着重要作用,尤其是维吾尔族人群TGF-β1基因多态性引起GDM易感性情况更严重。 Objective To analyze the relationship between TGF-β1 gene polymorphism and the susceptibility to gestational diabetes mellitus(GDM) in Uygur and Han nationalities of Xinjiang Autonomous Region. Methods Totally 450 cases of pregnant women admitted to our hospital from June 2014 to December 2017 were selected and divided into GDM group(n = 200,diagnosed as GDM,each 100 cases in Uygur and Han nationalities) and control group(n = 250,normal pregnant women,100 cases of Han nationality and 150 cases of Uygur nationality) according to the GDM diagnosis standard. The level of TGF-β1 was compared between the two groups,and the single nucleotide polymorphisms(SNPs) of TGF-β1 gene were determined and analyzed by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) :-509 C/T,+ 869 T/C and + 915 G/C. Results The level of serum TGF-β1 in GDM group was higher than that in control group,and the level of TGF-β1 in the Uygur nationality was higher than that in the Han nationality in GDM group(P 〈0. 05). The frequencies of CC,CT and TT genotypes at-509 C/T locus and the frequencies of GG,GC and CC genotypes at + 915 G/C locus were not significantly different between the two groups(P〉 0. 05),but the frequencies of TT,TC and CC genotypes at + 869 T/C locus were significantly different between GDM group and control group(P〈 0. 01). The frequencies of T and C alleles at + 869 T/C in GDM group were significantly different from those in control group(P 〈0. 01). The frequencies of TT,TC and CC genotypes and alleles(T,C) at + 869 T/C locus in the Uygur nationality in GDM group were significantly different from those in the Han nationality(P 〈0. 01). Conclusion The single nucleotide polymorphism of TGF-β1 at + 869 T/C locus may play an important role in the susceptibility to gestational diabetes mellitus in Uygur and Han nationalities of Xinjiang Autonomous Region,especially the susceptibility to GDM caused by TGF-β1 gene polymorphism is more serious in Uygur nationality.
