摘要
目的评价外源性雌二醇对更年期小鼠大脑神经元凋亡的保护作用。方法以C57/BL6小鼠为实验对象,通过双侧卵巢切除术制作更年期小鼠模型。实验动物随机分为对照组、假手术组、手术组、手术+雌二醇组,其中手术+雌二醇组自卵巢去势术后第6周开始隔日给予皮下注射雌激素0.5 mg·kg-1持续2周。ELISA法测定血清雌二醇水平;免疫组化法、Western blot法检测大脑组织中凋亡诱导因子的表达。结果手术组血清雌二醇水平低于假手术组,手术组凋亡诱导因子(apotosis inducing factor,AIF)表达量明显高于假手术组,手术+雌二醇组AIF表达量明显低于手术组。结论体内雌激素的下降能够促进小鼠大脑神经元的凋亡,补充雌二醇能够减缓更年期小鼠大脑神经元的凋亡。
Objective To evaluate the protective effect of exogenous estradiol on neuronal apotosis in climacteric mice. Methods To make menopausal mouse model,C57/BL6 mice were bilaterally ovariectomized.Experimental mice were randomly grouped into control group,sham-operated group,ovariectomized group,and ovariectomized plus estradiol-treated group.Six weeks after bilaterally ovariectomized operation,ovariectomized plus estradiol-treated group were subcutaneously injected estrogen 0.5 mg · kg -1 every other day for 2 weeks.The serum levels of estrogen were determined by ELISA,Immunohistochemistry and Western blot were used to evaluate the expression of apotosis-inducing factor(AIF)in brain tissue. Results Compared with sham-operated group,the levels of serum estrogen in ovariectomized group were decreased and the expressions of AIF were increased.The expression of AIF in ovariectomized plus estradiol-treated group was lower than that in ovariectomized group. Conclusion The deficiency of estrogen can induce the apotosis in mice brain neurons,and exogenous estradiol can slow down the apotosis of neurons in menopausal mice.
