摘要
【目的】通过不同浓度苯甲酸钠对斑马鱼胚胎发育的影响,初步探讨苯甲酸钠的毒理性质及对生物产生的影响。【方法】采用不同浓度的苯甲酸钠培养液,用于斑马鱼胚胎的发育影响,观察,统计孵化率、孵出仔鱼畸形率、死亡率等生理指标。【结果】斑马鱼直接暴露在苯甲酸钠溶液中,8h后高浓度组(10000mg·L^(-1))会造成外包层变薄,16h后实验组卵黄形状不规则,24h后实验组已出现黑色素的沉积,浓度大于1000mg·L^(-1)的实验组存在尾部发育不全、脊柱弯曲的个体,随苯甲酸钠的浓度升高马鱼胚胎死亡率上升,3天后死亡率,高浓度组达30.5%,畸形率9%。与对照组有显著性差异(P<0.05)。【结论】低浓度的苯甲酸钠对斑马鱼胚胎发育无显著影响,但高浓度(浓度大于1000mg·L^(-1)时)会导致胚胎发生黑色素沉积、个体发育不全等畸变。
Objective : to preliminary discuss and explore the toxicological properties of Sodium benzoate and its effects on biological production by studying the effects of different concentrations of Sodium benzoateon zebrafish embryo development. Method : differentconcentrationsofsodium benzoate culture medium were used for the development of zebrafish embryos, and the physiological indexes such as hatching rate, hatching larval rate and mortality were observed and recorded. Result : The zebrafish was exposed directly to sodium benzoate solution. After 8 h, the outer layer became thinner at high concentration( 10000 mg·L^(-1)). After 16 h, the shape of yolk in experimental group was irregular. After24 h, the deposition of melanin in experimental group was more than In the 1000 mg·L^(-1) experimental group, there was tail hypoplasia and spinal curvature. The embryo mortality of the fish increased with the concentration of sodium benzoate. After 3 days, the mortality rate was 30. 5 % in the high concentration group and the rateof deformity was9 %. There was significant difference with the control group( P〈0. 05). Conclusion : Sodium benzoate at low concentration has little effect on zebrafish embryos, but high concentrations( at concentrations above 1000 mg ·L^(-1)) lead to embryonic aberrations.
