鼠妇粉酶解最佳用酶的初步筛选 被引量:3

摘要 用胰蛋白酶、胃蛋白酶、胰凝乳蛋白酶、弹性蛋白酶四种酶对鼠妇粉进行酶解并选出最佳用酶。利用四种酶分别酶解鼠妇粉,通过观察乳腺癌MCF-7细胞的抑制率以及蛋白质的水解度,作为实验评价指标,综合评价四种酶对于鼠妇粉的酶解效果,最终甄选出最佳酶解用酶。实验测得胃蛋白酶酶解液水解度为19.36%,胰凝乳蛋白酶酶解液水解度19.97%,胰蛋白酶酶解液水解度为34.70%,弹性蛋白酶水解度分别为34.90%。体外抗肿瘤试验表明,在浓度为0~2 mg·mL^(-1)范围内,胰蛋白酶酶解液对乳腺癌MCF-7细胞抑制效果最佳,胃蛋白酶酶解液抑制效果更低,未酶解的抑制率最低,几乎无效。当浓度低于0.7 mg·mL^(-1)时,弹性蛋白酶水解提取液抑制率高于胰凝乳蛋白酶水解提取液的抑制率,当浓度在0.7~2 mg·mL^(-1)时,两种酶的抑制作用恰好相反。鼠妇粉酶解工艺最佳用酶为胰蛋白酶,胃蛋白酶效果最差,弹性蛋白酶和胰凝乳蛋白酶效果介于二者之间。 The enzymolysis of pillworm powder was carried out by respectively using pepsin,elastase,trypsin and chymotrypsin,and the best one was selected. The pillworm powder was respectively hydrolyzed by the four enzymes, the hydrolysis degree of the protein and the cell inhibition rate of breast cancer MCF-7 were used as the index to evaluate them. The hydrolysis degrees of trypsin, pepsin, chymotrypsin and elastase were 34.70%, 19.36%, 19.97% and 34.90%, respectively. In vitro antitumor test showed that trypsin hydrolysis effect was the best,pepsin hydrolysis effect was lower than the others, and the pillworm without enzymolysis nearly had no effect on the breast cancer MCF-7 cells when the concentration was in the range of 0 to 2 mg·mL^(-1), when the concentration was less than 0.7 mg·mL^(-1), the inhibitory rate of elastase hydrolysis extract was better than chymotrypsin, when the concentration was 0.7~2 mg·mL^(-1), the inhibition of these two enzymes was just the opposite. The optimal enzyme for enzymolysis of pillworm powder was trypsin, followed by elastase and chymotrypsin, and pepsin was the worst.
