摘要
目的探讨肝内胆管癌(ICC)中野生型p53诱导的蛋白磷酸酶1(Wip1)、基质金属蛋白酶-2(MMP-2)表达水平及临床意义。方法选取38例ICC患者肝内胆管癌组织标本为研究对象,同期选取36例正常胆管组织标本为对照,采用免疫组织化学(S-P)方法检测组织中Wip1、MMP-2蛋白表达水平,分析二者与ICC临床病理特征之间的关系。结果 ICC组织中Wip1蛋白阳性表达率为78.95%,明显高于正常胆管组织中Wip1蛋白阳性表达率8.33%,差异有统计学意义(P<0.05);ICC组织中MMP-2蛋白阳性表达率为73.68%,明显高于正常胆管组织中MMP-2蛋白阳性表达率5.56%(P<0.05)。Wip1蛋白表达与ICC患者年龄、性别、临床分期、有无肝炎、组织学分级无明显相关性(P>0.05),与患者门静脉浸润、淋巴结转移密切相关(P<0.05);MMP-2蛋白与ICC患者年龄、性别、有无肝炎、组织学分级无明显相关性(P>0.05);与患者临床分期、门静脉浸润、淋巴结转移密切相关(P<0.05)。Wip1与MMP-2在ICC组织中表达呈较强正相关(γ_s=0.683,P<0.05)。结论 Wip1、MMP-2在ICC中明显高表达,二者参与ICC发生及发展。
Objective To investigate the expression and clinical significance of Wip1 and MMP-2 in Intrahepatic cholangiocarcinoma. Methods A total of 38 ICC tissue samples were collected,and 36 normal bile duct tissues were selected as control. The expression of Wip1 and MMP-2 protein in tissues was detected by S-P,the relationship between the two and the clinicopathological features of ICC were analyzed. Result The positive expression rate of Wip1 protein in ICC tissues was 78. 95%,which was significantly higher than the normal bile duct tissue 8. 33%. The positive expression rate of MMP-2 protein in ICC tissues was 73. 68%,which was significantly higher than the normal bile duct tissues 5. 56%( P 〈 0. 05). There was no significant correlation between Wip1 protein expression and ICC patients’ age,sex,clinical stage,hepatitis and histological grade( P 〉 0. 05),which was closely related to portal vein infiltration and lymph node metastasis( P 〈 0. 05). There was no significant correlation between MMP-2 protein and ICC patients’ age,sex,hepatitis and histological grade( P 〉 0. 05),which was closely related to clinical stage,portal vein infiltration and lymph node metastasis( P 〈 0. 05). There was a strong positive correlation between Wip1 and MMP-2 expression in ICC tissues( γ_s= 0. 683,P 〈 0. 05). Conclusion Wip1,MMP-2 have significantly higher expression in ICC,and are involved in ICC occurrence and development.
