环介导等温扩增技术快速检测鲜切蔬菜中的大肠杆菌

摘要 为鲜切蔬菜快速检测的市场化应用提供理论基础,利用大肠杆菌的特异性基因rfbE基因设计LAMP引物,通过对10株菌株进行特异性检测,建立利用环介导等温扩增技术检测鲜切蔬菜中的大肠杆菌O157∶H7的方法,并进行验证检测。结果表明:检测鲜切蔬菜样品特异性为93.3%,准确度为96.5%,检测大肠杆菌灵敏度达1.69CFU/mL。建立的快速检测方法可满足市场化应用的需要。 In order to provide a theoretical basis for market application of rapid detection of fresh-cut vegetables,the loop-mediated isothermal amplification(LAMP)primer was designed with the specific gene rfbEof Escherichia coli,specific detection of 10 strains was conducted to establish the detection method of E.coli O157∶H7 in fresh-cut vegetables by LAMP,then the detection results were verified as well.Results:The specificity of fresh-cut vegetables was 93.3% with the accuracy of 96.5%,and the detecting sensitivity of E.coli was up to 1.69 CFU/mL.The established rapid detection method could meet the requirement of market application.
