高氧液抑制根管内常见厌氧菌的研究 被引量:3

摘要 目的:探讨高氧液作为根管冲洗剂对厌氧菌的作用效果。方法:制备高氧液;取变异链球菌、中间普氏菌、粘性放线菌培养48h后,制成1×10~6CFU/mL的菌悬液;将实验分为高氧液组、5g/LNaClO组、生理盐水3组,分别对3种厌氧菌进行菌落形成实验、抑菌环实验,并纳入慢性根尖周炎患者82例共96个患牙分为3组(n=32)进行根管冲洗效果比较,RT-PCR初步探讨高氧液抑菌的作用机制。结果:制备的高氧液氧分压为(106.9±5.3)kPa;高氧液组和5g/LNaClO组处理细菌悬液后,3种厌氧菌的菌落形成数均少于生理盐水组(P<0.05),而抑菌环均显著大于生理盐水组(P<0.05)。分别使用高氧液、5g/LNaClO和生理盐水冲洗根管后,高氧液组和5g/LNaClO组根管内残余厌氧菌的菌落数均显著少于生理盐水组(P<0.05)。与需氧的金黄色葡萄球菌比较,厌氧的变异链球菌、中间普氏菌、粘性放线菌的SOD基因表达水平均较低(P<0.05);与生理盐水相比,高氧液组3种厌氧菌中Caspase3基因的表达显著上调(P<0.05)。结论:高氧液对根管内常见的3种厌氧菌具有抑菌作用,其原理可能是促进细菌的程序性死亡。 AIM: To study the effects of hyperoxia solution (HS) as a flushing agent against S.mutans, P.intermedia and A.viscosus in the process of root canal preparation. METHODS: HS was prepared and was used in the cultrde of bacteria. Colony formation experiment and the bacteriostasis loop experiment were carried out for the examination of bacteria proliferation. Then 96 teeth with chronic periapical periodontitis in 82 subjects were irrigated by HS (HS group), 5 g/L sodium hypochlorite (SH group) and normal saline (control group) respectively ( n =32), the effects were compared among groups. Finally, the possible mechanism of the bacteriostasis of HS was studied by RT-PCR. SPSS 19.0 software was used for data analysis. RESULTS: The oxygen partial pressure of the HS was(106.9±5.3)kPa, and the oxygen concentration was 5.2 times as high as that of the conventional liquid. The number of colony formation of 3 bacteria in the HS group and the SH group were less than that in the control group ( P 〈0.05), the bacteriostasis ring was significantly greater than that of the control group ( P 〈0.05). The in vivo study showed that the anaerobe bacteria in the HS group and the SH group were significantly less than that in the control group ( P 〈0.05). RT-PCR examination showed that, compared with the aerobic staphylococcus aureus, the SOD gene expression of S. mutans, A. viscosus and P. intermedia were lower in HS group( P 〈0.05). HS up-regulated the expression of Caspase3 gene in the three anaerobes ( P 〈0.05 ). CONCLUSION: Hyperoxia solution may promote the apoptosis of bacteria by oxygen toxicity and thus inhibit the three kinds of anaerobes in the root canal.
