脐带、脂肪源性间充质干细胞改善肝硬化大鼠的效果比较 被引量:3

摘要 目的比较脐带间充质干细胞(umbilical cord mesenchymal stem cells,UC-MSCs)和脂肪源性间充质干细胞(adipose-derived mesenchymal stem cells,ADSCs)对肝硬化大鼠的治疗效果。方法四氯化碳(CCl4)诱导大鼠肝硬化模型。成模大鼠随机分为UC-MSCs组、ADSCs组和对照组,每组10只,分别注入UC-MSCs、ADSCs各2 ml(细胞数量5×106)及等量生理盐水。干细胞移植前及移植4周后,检测大鼠肝功能;取肝组织制备切片进行HE、Masson染色;免疫组化法检测肝脏中α-平滑肌肌动蛋白(α-SMA)的表达。结果干细胞移植后,UC-MSCs组和ADSCs组的肝功能较对照组有明显改善(P<0.05)。组织病理提示,细胞移植组较对照组在肝组织炎症活动度和纤维化程度评分等方面均有明显降低,差异均有统计学意义(P<0.05);UC-MSCs组与ADSCs组间肝组织评分差异无明显统计学意义(P>0.05)。UC-MSCs组、ADSCs组与对照组相比,α-SMA表达差异有统计学意义(P<0.05);UCMSCs组与ADSCs组α-SMA表达差异无统计学意义(P>0.05)。结论 UC-MSCs、ADSCs移植能改善肝硬化大鼠的肝功能和病理组织学,两者效果相似。 Objective To compare the therapeutic effect of umbilical cord mesenchymal stem cells( UC-MSCs) and adipose-derived mesenchymal stem cells( ADSCs) on hepatic cirrhosis of rats. Methods Rat model of hepatic cirrhosis were induced by carbon tetrachloride( CCl4). The rats were randomly divided into UC-MSCs group,ADSCs group and control group,10 rats in each group,which were injected with UC-MSCs,ADSCs 2 ml( 5 × 106 cells),and equal volume of saline,respectively. The liver function was measured before and after 4 weeks of transplantation. HE and Masson staining were performed on the liver tissue. The expression of α-smooth muscle actin( α-SMA) in the liver was detected by immunohistochemistry. Results After cell transplantation, the liver function of UC-MSCs group and ADSCs group were significantly improved compared with the control group ( P 〈0.05). Tissue and pathology suggested that the activity index of inflammation and degree of fibrosis in UC-MSCs group and ADSCs group were significantly lower than those in control group, the difference was statistically significant ( P 〈0.05). However, there was no significant difference in liver tissue score between UC-MSCs group and ADSCs group ( P 〉0.05). Compared with control group, α-SMA expression in UC-MSCs group and ADSCs group was signilicantly different; there was no significant difference in α-SMA expression between UC-MSCs group and ADSCs group ( P 〉0.05).Conclusion UC-MSCs and ADSCs transplatation can improve liver function and disease histopathology of cirrhotic rats,and these two approaches have comparable clinical effects,and the effect is similar.
